Abstract: PURPOSE:To quickly determine intracellular ATP for microbial examination, etc., in high sensitivity without diluting the specimen by contacting an extracted specimen of intracellular ATP with an agent for suppressing the inhibition of enzymatic reaction and containing cyclodextrin and applying a biochemical luminescent method to the contact product. CONSTITUTION:ATP in a cell is determined in high sensitivity for food sanitation, clinical examination, environmental microbial examination, etc., by contacting a specimen containing cells of microorganisms, etc., with an adenosine triphosphate (ATP) extraction reagent containing a surfactant (e.g. dodecyldimethylbenzyl ammonium chloride) to extract the intracellular ATP of the specimen, contacting the extracted specimen with an agent for suppressing the inhibition of enzymatic reaction and containing a cyclodextrin (alpha- cyclodextrin), adding a luminescent reagent containing luciferin as a fluorescent substrate and luciferase as a luminescent enzyme to the extracted specimen to apply biochemical luminescent method and measuring the light emitted by the biochemical luminescence caused by the enzymatic reaction of luciferin, luciferase and ATP.

Abstract: PURPOSE:To provide a method and apparatus for measuring ATP quantity of cell which enables the ATP (adenosine triphosphate) quantity of cells to be measured with high precision dispensing with the operation of folding a filter membrane after cell trap and not only improving operability but also solving the dangerousness of contamination from outside the filter membrane. CONSTITUTION:A filter membrane 100 which has trapped cells in a specimen is placed in a one-sided open tray container 200 without being folded. The container 200 is injected with an extraction reagent 104 and a luminous reagent to cause biochemical luminous reaction. A photomultiplier tube 108 is installed so as to face the opening of this container 200 to detect the luminous quantity, resulting in measurement of the ATP quantity of cells.

Abstract: PURPOSE:To provide a chemical light emission detector, wherein the detection of light emission and emitted light quantity with an optical detecting means is done at a constant temperature, an measurement of emitted light quantity due to light emission reaction is done with high precision, stability and reliability with no affect from temperature. CONSTITUTION:A light-shielding case 1 that contains a flaw-cell 2 and a photomultiplier tube 3 is provided in a heat-insulation case 11, and further, a constant- temperature equipment 7 that contains a heater and a cooler is also provided in it. The constant-temperature equipment 7 keeps the atmosphere in the case 1 at a specified temperature, or constant temperature, so the detection of light emission reaction and emitted light quantity of a measured sample, introduced in the flaw-cell 2, with the photomultiplier tube 3, is done at a constant temperature, thus objective is attained.

Abstract: PURPOSE:To provide an in-cell ATP measuring device that allows measuring of ATP amount of both somatic cell and microorganism cell with ease and in a short time without causing error attributable to operators by performing automatic preprocessing and extraction operations with an object sample instead of manual operation by an operator. CONSTITUTION:An entire cell extraction cell 28, a somatic cell extraction cell 29, a means that supplies these cells 28 and 29 with a cell suspended liquid 5, a means that injects entire cell extraction reagent 3 and somatic cell extraction reagent 4 into the supplied cell suspended liquid 5, a means that injects light emission reagent 1 into the obtained entire cell extraction reagent and somatic cell extraction, a chemical light emission detector 30 that detects amount of light emission of the entire cell extraction reagent and somatic cell extraction reagent, and a computer 32 are provided, and the computer 32 controls operation of respective parts. With this, manual operation by an operator is not required, so objective is attained.
