Abstract:
본 발명은 SET/TAF-Iβ 및 Ku70/80 결합을 이용한 항암제 스크리닝 방법에 관한 것으로서, 상세하게는 SET/TAF-Iβ 및 Ku70/80 결합을 촉진하는 후보약물을 선별하는 단계를 포함하는 항암제 스크리닝 방법을 제공한다. 한편, INHAT 복합체의 서브유닛인 SET/TAF-1β가 Ku70/80과 결합하고, 상기 결합이 CBP- 및 PCAF-매개 Ku70 아세틸화를 억제한다는 것을 확인하고, DNA가 손상되면 SET/TAF-1β와 Ku70/80의 결합을 방해하므로, 상기 결합을 이용하여 DNA 손상과 관련된 질환인 암을 치료할 수 있는 항암제 스크리닝 방법으로 유용하게 사용될 수 있다.

본 발명은 JMJD1B (jumonji domain containing 1B)유전자, 이의 전사체 또는 발현된 단백질을 포함한 백혈병 진단용 조성물을 제공한다. 보다 구체적으로, 상기 JMJD1B의 전사, 단백질 발현 또는 단백질 활성을 억제하면 암 유전자의 활성이 억제되고 세포자살 유전자를 활성화시켜 백혈병 치료에 효과가 있으므로, 상기 유전자, 유전자의 전사나 발현, 활성을 조절함으로 백혈병 진단제, 약물 스크리닝 등이 가능하고, 본 발명의 유전자를 백혈병 치료 타겟으로 활용할 수 있을 것이다.

PURPOSE: A composition containing JMJD1B gene, a transcript thereof, or proteins expressed from the same is provided to suppress oncogene activation and to treat leukemia. CONSTITUTION: A composition for diagnosing leukemia contains JMJD1B(jumonji domain containing 1B) encoding an amino acid of sequence number 1, a transcript thereof, or protein expressed from the same. The JMJD1B protein has histone dimethylase H3K9-me1/2 activity. A method for screening a therapeutic agent for treating leukemia comprises a step of contacting the gene, transcript, or protein as a target material with a test material; a step of identifying a reaction of the target material and the test material; and a step of determining the gene transcription or protein expression or activation.

CONSTITUTION: A method for inducing cell apoptosis comprises a step of overexpressing proteins containing SET domains of amino acid sequence number 1. The proteins are setd3 of sequence number 2. A pharmaceutical composition for inducing cell apoptosis contains the proteins or a gene encoding the same as an active ingredient. The pharmaceutical composition is used for preventing and treating cancer, diabetes, artheriosclerosis, inflammatory bowel diseases, autoimmune diseases, transplantation rejection, post-traumatic immune response, infectious diseases, lymphatic tumor, multiple sclerosis, SLE, pachydermia, ischemia, stroke, uveitis or diabetic retinopathy.

An anti-cancer composition containing DEK proteins or nucleic acid coding the DEK proteins is provided to induce apoptosis and inhibit dysplasia of cancer cells by overexpressing DEK to increase caspase-dependent apoptosis. An anti-cancer composition for inducing apoptosis comprises DEK proteins having amino acid sequence of SEQ ID NO:2 or nucleic acid for coding DEK protein having base sequence of SEQ ID NO:1. A transformed drosophila is prepared to overexpress the DEK proteins. The transformed drosophila is obtained by a pUAS-hDEK recombinant vector containing the nucleic acid for coding the DEK proteins. The anti-cancer composition containing the DEK proteins or the nucleic acid coding the DEK proteins is used for treating or preventing cancer.

A nucleic acid for leukemia diagnosis comprises base sequence of SEQ ID NO:1 or complementary base sequence thereof or can be specifically coupled with the base sequence of SEQ ID NO:1 or the complementary base sequence thereof. An oligonucleotide chip for leukemia diagnosis contains the nucleic acid for leukemia diagnosis. A method for diagnosing leukemia has a step of measuring existence of expression of RE-IIBP(response element-II binding protein) in monocyte of marrow or measuring existence of methylation of histone protein in the monocyte of marrow.
