Abstract: PURPOSE: A method for preparing a hyaluronic acid-protein conjugate is provided to control aldehyde substitution rate of hyaluronic acid-aldekyde derivatives. CONSTITUTION: A method for preparing a hyaluronic acid-protein conjugate comprises a step of reacting protein N-terminal with hyaluronic acid-aldehyde derivative in which an aldehyde group is introduced in hyaluronic acid or salt thereof. The hyaluronic acid-aldehyde derivative is prepared by reaction of hyaluronic acid or salt thereof with oxidizer. The oxidizer is periodate. A reagent which induces reduction amination is sodium cyannoborohydride(NaBH_3CN) or sodium triacetoxyborohydride(NaBH(OCOCH_3)_3).
