Abstract: PURPOSE: An in vitro kit for quantitative measurement of protease activity and a method for manufacturing the same are provided to specifically emit strong fluorescence. CONSTITUTION: An in vitro kit for quantitative measurement of protease activity comprises a complex in which fluorescence and quencher are combined and a peptide substrate is conjugated to a polymer. The complex has structural formula 1. The quencher is a blackhole quencher, blackberry quencher, or derivative thereof. A method for manufacturing the kit comprises: a first step of synthesizing a specific peptide substrate to a specific protease and reacting with near infrared ray fluorescence; a second step of binding the peptide substrate with quencher; a third step of binding polymers to fluorescence-peptide substrate-quencher; and a fourth step of fixing the fluorescence-peptide substrate-quencher-polymers onto a 96-well.

Abstract: 본발명은항암상승효과를나타내는종양세포특이적자기조립나노약물복합체에관한것으로, 보다구체적으로는종양세포에존재하는효소에의해서분해되어, 활성성분인항암약물및 약물내성억제펩타이드(SMAC)를동시에방출할수 있는나노약물복합체에관한것이다. 본발명에따른약물복합체또는자기조립나노입자는종양조직에서특이적으로과발현하는카텝신 B 효소에의해분해되어, 독소루비신및 SMAC을방출하는프로드러그(prodrug)로, 종양세포에특이적으로반응하여활성화되기때문에, 암예방또는치료과정에서유도되는심각한정상조직손상및 독성등의부작용을해결할수 있다. 또한, 본발명에따른약물복합체또는자기조립나노입자는수성상태에서추가적인고분자및 전달체없이안정한나노입자를형성하므로기존의고분자및 전달체기반의약물전달제형보다높은약물봉입및 전달효율, 대량합성및 QC가용이하다. 또한, 본발명에따른약물복합체또는자기조립나노입자는독소루비신및 SMAC을암 세포에방출하여상승효과에의해우수한항암효능을나타낼뿐만아니라, 기존항암화학치료의문제였던약물내성을극복할수 있다.

Abstract: 본발명은표피생장인자수용체 (EGFR)-양성종양표적화소광나노프로브및 이의제조방법에관한것으로서, 더욱구체적으로는, 표피생장인자 (EGF)에, 형광체및 소광체가컨주게이션된것을특징으로하는표피생장인자수용체 (EGFR)-양성종양표적화소광나노프로브및 이의제조방법에관한것이다. 본발명에따른표피생장인자수용체 (EGFR)-양성종양표적화소광나노프로브 (EGF-NP)는표피생장인자수용체-양성종양을특이적으로검출할수 있으며, 따라서표피생장인자수용체-표적화된약물의치료효과를모니터링하는데매우효과적이다. 또한, 짧은영상화시간을갖기때문에, 반복적인영상획득을쉽게간단하게수행할수 있도록하며, 시간경과에따른연속적영상데이터를획득하는데유용하다. 더불어, 본발명에따른 EGF-NP는소정시간경과후 체내에서소멸되기때문에, 신체안전성측면에서도우수하다.

본 발명에 따른 복합체는 소광체의 형광물질에 대한 높은 소광 능력으로 형광이 발광하지 않은 상태로 있다가 특정 단백질 분해효소에 의하여 펩타이드 기질이 분해가 되어야만 강한 형광을 특이적으로 발광함으로써, 특히 단백질 분해효소 과다 발현을 억제하는 억제제와 같은 신약을 스크리닝하는 방법, 암, 골 관절염, 류마티스 관절염, 치매 등 자가면역질환 등의 다양한 질병 및 난치병의 조기 진단에 유용하게 사용될 수 있다.-
