    Abstract: PROBLEM TO BE SOLVED: To provide a method for producing a nitric ester safely on an industrial level by using only inexpensive reagents under mild reaction conditions without using a strong acid and a reaction or a reagent having high danger. SOLUTION: In a nitration reaction of a compound containing one or more hydroxyl group, nitric acid is added to a solution containing the compound, an acid anhydride, a carboxylic acid and an organic solvent. Preferably nitric acid is gradually dripped or dropped dividedly twice or more times. Preferably nitric acid is dripped on the solution containing the carboxylic acid, the organic solvent, the acid anhydride and the compound containing one or more hydroxyl groups and then the addition or drop of the compound containing one or more hydroxyl groups which may be dissolved in the organic solvent and the drop of nitric acid are successively carried out and, if required, repeatedly carried out.

    Abstract: PURPOSE:To obtain the titled compound useful as a raw material of acetoacetic acid 3-nitratopropyl ester which is a synthetic raw material of a compound useful as a remedy for circulatory diseaes, economically, by reducing a 3-nitratopropionic acid lower alkyl ester with an inexpensive reducing agent. CONSTITUTION:The titled compound can be produced by reducing 3-nitrato- propionic acid lower alkyl ester in an inert solvent with a borohydride or aluminum hydride reducing agent. The temperature of the reaction is 20-120 deg.C for borohydride-type reducing agent, and -50-+30 deg.C for aluminum hydride-type reducing agent. The most preferable reducing agent is NaBH4-CaCl2, and the most preferable 3-nitratopropionic acid lower alkyl ester is methyl 3-nitratopropionate. The inert solvent is e.g. methanol, dimethoxyethane, etc. for the borohydride-type agent and ether, etc. for the aluminum hydride-type agent.

    Abstract: PURPOSE:The titled stable and easily handleable inclusion compound useful as a drug, capable of relieving irritation and preventing volatility, having high safety for tableting, molding, etc., obtained by including an isosorbidnitrate with cyclodextrin. CONSTITUTION:An isosorbidnitrate having a coronary blood vasodilating action, a dinitro ester or mononitro ester derivative of isosorbidnitrate widely used as a remedy and preventive for attack of angine pectoris in the form of a sublingual tablet, oral drug, sustained release drug, injection, etc., is dissolved or dispersed into a solvent containing preferably an equimolar amount of cyclodextrin and water, and the solvent is eliminated to give an isosorbidnitrate- cyclodextrin inclusion compound. alpha-, beta- and gamma-cyclodextrin, a cyclic oligosaccharide called cycloamylose, etc., comprising 6-8 constitutional units of D- glucopyranoses is used as the cyclodextrin.
