Abstract: PURPOSE:To obtain fine particles for agglutination, used for the measurement of the immune composite bodies in a body fluid, which are sensitive and excellent both in maintaining a constant quality and in the preservation stability, by fixing conkurchinine to individually separated fine particles whose particle sizes are within a specified range by means of a physical absorption or chemical bonding. CONSTITUTION:Conkurchinine is physically absorved or chemically bonded to a hydrophobic macromolecular compound, the erythrocyte of an animal which has been treated with formalin and moreover treated with tanic acid or the like, and fine particle carriers of a hydrophilic insoluble substance or the like which has a functional group capable of chemically bonding conkurchinine by means of covalent or ionic bonding. As said fine particles, fine particles of average particle size 0.01- 20mu and individually separated are used. Specially, the fine particles covalent-bonded to a hydrophilic macromolecular compound hardly cause a nonspecific agglutination. The dispersion of sid fine particles and a body fluid are reacted with each other to measure the settling image resulting from an agglutination. Thereby, the measurement of the immune composite bodies in the body fluid is readily performed with high sensitivity.

Abstract: PURPOSE:To obtain the titled fine particles for clinical tests having a good measuring sensitivity, by polymerizing acrylonitrile or methacrylonitrile into the form of fine particles, hydrolyzing the fine particles, and reacting an immunological active substance with the resultant specific carrier particles. CONSTITUTION:Acrylonitrile or methacrylonitrile is polymerized in a medium, e.g. ethyl propionate, which will dissolve the monomer mixture but precipitate and deposit the resultant polymer in the presence of a crosslinkable monomer, e.g. divinylbenzene, having at least two polymerizable carbon-carbon double bonds to precipitate and deposit a fine particulate polymer having an average diameter of 0.03-10mum. The resultant fine particulate polymer is then hydrolyzed and reacted with an immunologically active substance, e.g. syphilitic treponemal antigen or HBs antigen, to give immunologically active fine particles. The resultant carrier consists of polyacrylamide particles having a uniform particle size, and has very little nonspecific adsorption, e.g. protein, and facilitates the measurement.

Abstract: PURPOSE: To enable an immunoassay of high sensitivity by enhancing the light emission reaction of cypridina luciferase in the presence of calcium ion and using the cypridina luciferase as a marker for immunoassay reagents. CONSTITUTION: Cypridina luciferin as a light emission substrate is dissolved in 10mM sodium citrate buffer.140mM sodium chloride solution (pH4.5) and calcium ion such as calcium chloride is added to the solution in an amount of 5μM-5 mM. Then, A 100mM tris hydrochloride buffer solution containing cypridina luciferase (pH7.2) is added to the luciferin solution and, after 10 seconds, the 10-second emission is measured with a luminometer. This measurement process is carried out in the presence of calcium ion to increase both of the emission value and the S/N ratio to increase the measurement sensitivity more than double in comparison with the luciferin measurement without calcium ion.

Abstract: PURPOSE:To reduce nonsingular adsorption and to facilitate operation and shorten its required time by separating and removing a labeled material which does not form a composite body by electrophoresis and detecting the remaining labeled material which forms the composite body. CONSTITUTION:A flat plate 1 having many through holes 2 for holding a sample or gel and a conductive solution layer 5 for the electrophoresis of a sample layer 4 and a gel layer 3, and the solution layer 5 consists of two liquid layers having electrodes 6 and 6' with the mutually opposite polarities for the electrophoresis. Then samples are collected in plural reaction containers and a labeled material is added and made to react for producing the composite body. The gel layer 3 is prepared in the holes 2 simultaneously. Then the sample layer 4 is superposed on the gel layer 3 and the flat plate 1 is so arranged that its reverse surface, i.e. gel layer 3 contacts the solution surface of the solution layer 5. Further, the conductive solution is injected into the solution layer 5 on the top surface of the flat late 1 and then a specific voltage is applied for a specific time. Consequently, the labeled material which does not form the composite body is separated and removed by electrophoresis, so the labeled material of the composite body which remains in the gel can be detected.

Abstract: PURPOSE:To obtain fine particulate polymer having an orderly particle diameter distribution, by charging a liquid chamber provided with a piezoelectric element with a monomer, increasing the pressure in this chamber by applying a pulsed voltage to the element, discharging a fine particulate monomer from a nozzle and polymerizing and curing the monomer. CONSTITUTION:The front end of a liquid chamber 20 communicating with a feed tank 10 is provided with a nozzle 21 having fine holes. Liquid chamber 20 is surrounded with a piezoelectric element 22. After liquid chamber 20 has been filled to its front end with a monomer or an oligomer fed from the feed tank 10, a pulsed voltage from a means 23 for applying a pulsed voltage is applied to piezoelectric element 22 to allow it to contract inwardly as shown by the broken lines and generate a pressure in liquid chamber 20. In this way, fine particulate monomer or oligomer is discharged from nozzle 21 and is received by a receiver 30 filled with a liquid in which the monomer or the oligomer is not soluble. This monomer or oligomer is polymerized and cured to obtain fine particulate polymer.
