Abstract:
The present invention relates to a polypeptide based block copolymer having biodegradability by peptidase, a method for preparing the same, and polymeric micelles using the same. The block copolymer according to the present invention is a block copolymer of: a polyethylene glycol based compound of which the solubility to water varies depending on pH but which does not form micelles through self-assembling; and a polyglutamic acid based compound which is formed by an aminolysis reaction of glutamic acid and tertiary amine of which one end of an alkyl group is substituted with NH_2 or an aminolysis reaction of triamine and glutamic acid. The block copolymer of the present invention retains pH sensitivity as well as biodegradability by in vivo peptidase, and thus its degrading rate associated with the drug release period can be controlled. Therefore, a target-oriented drug delivery system dependent on the pH change and a drug delivery system capable of controlling a degrading rate can be provided.

본 발명은 질환의 진단 및 치료를 위한 다양한 진단제와 다양한 치료제 전달을 위한 전달체로서 사용 가능할 수 있는 pH 민감성 공중합체, 이의 제조방법 및 이를 이용한 폴리머좀(polymersome) 약물 조성물에 관한 것으로서, 보다 상세하게는 pH에 따라 물에 대한 용해도 특성을 갖는 폴리(β-아미노 에스터) 화합물에 원래 가지고 있는 pH 민감성기외에 측쇄로 하이드록실기를 갖도록 분자설계한 후에 락타이드를 개환중합을 통해 공중합시켜 소수성 블록을 형성시킨 후에 친수성 블록으로말단이 카르복실기로 치환된 폴리에틸렌글리콜 화합물을 공중합시킴으로써, 소수성 및 친수성 약물 양쪽다 전달이 가능한 pH 민감성 폴리(β-아미노 에스터)-폴리락틱산-폴리에틸렌글리콜 공중합체 및 이의 제조방법, 상기 공중합체를 포함하는 폴리머좀형 진단제 및 약물전달을 위한 조성물을 제공하는 것이다.

PURPOSE: A drug delivery system for treating liver cancer based on interventional injection of temperature- and pH-sensitive hydrogel is provided to stably support an anticancer agent and to enable deep injection into microvessel. CONSTITUTION: A drug delivery system for treating liver cancer based on interventional injection of temperature- and pH-sensitive hydrogel contains a drug delivery system with a block copolymer and a therapeutic drug supported inside the drug delivery system. The drug delivery system further contains a contrast agent inside. The block copolymer is a poly(beta-amino ester)-based block copolymer, a polyamidoamine-based polymer, a polyaminoester urethane-based block copolymer, or a sulfonamide-based block copolymer. An injectable and biodegradable drug delivery system is provided to control glucose concentration in blood by slowly releasing insulin, improve compliance of patient by reducing injection frequency, and minimize side effects of drug by using polymer hydrogel. An injectable and biodegradable drug delivery system contains a negatively charged drug included in polymer hydrogel having a copolymer consisting of temperature sensitive and biodegradable hydrophilic polymer and hydrophobic polymer, coupled with a pH sensitive component in the end, wherein the drug becomes gel under conditions of pH 7.2-7.4 and 37 deg. C, and the pH sensitive component is ionized positively and forms an ionic complex with the negatively charged drug. Further, the hydrophilic polymer is one or more selected from a group consisting of polyethylene glycol, poly(N-2-(hydroxypropyl)methacrylamide), poly(divinyl ether-co-maleic anhydride), poly(styrene-co-maleic anhydride), or a polymer synthesized from a group consisting of dextran, chitosan, glycol chitosan, poly-L-lysine, polyglutamic acid and polyaspartic acid.

A temperature and pH sensitive block copolymer, its preparation method, and a polymer hydrogel type drug delivery vehicle containing the copolymer are provided to improve gel strength in case of injection, thereby inhibiting the initial over release of drug. A temperature and pH sensitive block copolymer comprises a copolymer of a polyethylene-based compound and a biodegradable polymer; and a polyurethane-based oligomer. Preferably the polyethylene-based compound is represented by RO-(CH2-CH2-O)n-H and has a number average molecular weight of 1,000-5,000 g/mol, wherein R is H or a C1-C5 alkyl group; and n is an integer of 11-45. Preferably the biodegradable polymer is a biodegradable aliphatic polyester; and the polyurethane-based oligomer is coupled to the copolymer.
