Abstract: Various light emitting diode (LED) based measurement systems and methods are provided. One system includes one or more arrays of LEDs arranged along a flow path of a sample. The array(s) are configured to illuminate the sample as the sample moves along the flow path. The system also includes one or more detectors configured to detect light resulting from illumination of the sample by the array(s). One method includes illuminating a microsphere at different positions along a flow path of the microsphere. The method also includes detecting light resulting from the illumination to produce individual output signals corresponding to the illumination at the different positions. The method further includes combining the individual output signals to produce a single output signal having a signal-to-noise ratio that is greater than a signal-to-noise ratio of the individual output signals.

Abstract: A system configured to acquire and process image data is disclosed. The system comprises an imaging subsystem configured to image, at different wavelength bands, particles disposed within the imaging subsystem and acquire data corresponding to emissions of imaged particles. The system further comprises a storage medium with program instructions which are executable by a processor for storing data acquired for multiple images of the particles, wherein each of the multiple images corresponds to a different wavelength band, selecting a reference image from one of the multiple images and manipulating the coordinates of at least one of the multiple images that is not the reference image such that spots corresponding to the particles within each of the multiple images align with spots in the reference image.

Abstract: Methods and systems for expanding the dynamic range of a system are provided . One method includes splitting fluorescent light emitted by a particle into multiple light paths having different intensities, detecting the fluorescent light in the multiple light paths with different channels to generate multip le signals, and determining which of the channels is operating in a linear rang e based on the multiple signals. The method also includes altering the signal generated by the channel operating in the linear rang to compensate for the different intensities. Another method includes illuminating a particle in multiple illumination zones with light having different intensities and separately detecting fluorescent light emitted by the particle while located in the multiple illumination zones to generate multiple signals. The method also includes determining which of the signals is located in a linear range and altering the signal located in the linear range to compensate for the different intensities.
