Abstract:
A production method for producing cell colony forming units in vitro from a mammalian tissue explant, the method comprises the steps of a) growing a piece of the mammalian tissue explant in a growth medium to obtain cell colony forming units from immature cells from the piece of explant, and b) harvesting cells from one or more of the cell colony forming units for use in Autologous Cell Implantation/transplantation methods. The mammalian tissue explant is selected from the group consisting of cartilage; bone such as, e.g., bone marrow; connective tissue; muscle tissue such as, e.g., smooth muscle tissue, heart tissue, liver tissue and skeletal muscle tissue; skin tissue such as, e.g., periosteum; mucosal tissue; brain tissue, pancreas tissue and blood vessels. In particularly, the mammalian tissue explant is cartilage, such as elastic, fibro, hyalin or articular hyalin cartilage. The cells obtained are suitable for use in autologous implantation/transplantation methods. In a specific embodiment, the cell obtained are chondroytes, especially for use in autologous chondrocyte implantation (ACI) methods.
Abstract:
The present invention provides for a method for tissue engineering by cell implantation that involves the use of a scaffold in situ at the site of a defect, where the therapeutic cells are fixed in place into the scaffold only once the scaffold is inserted at the site of the tissue defect, thereby locking not only the cells to the scaffold, but also the scaffold to the tissue defect. The invention also provides a kit of parts suitable for performing the method of the invention.

The present invention relates to an endoscopic method for treating cartilage or bone defects in a mammal, said method comprising the steps of: i.-dentifying the position of the defect, ii.-applying a cell suspension comprising cells selected from the group consisting of chondrocytes, chondroblasts, osteocytes and osteoblasts and combinations thereof into the cartilage or bone defect, the defect being positioned in such a manner that the cell suspension substantially does not flow out of the defect upon application.

An isolated, acellular biosynthetic cartilaginous matrix substantially devoid of synthetic biodegradable scaffold structure is provided. Through a method with the steps of a) contacting in vitro a population of chondrogenic cells with a synthetic biodegradable scaffold; b) culturing in vitro for a period of time said chondrogenic cells within said synthetic biodegradable scaffold so that the chondrogenic cells produce a biosynthetic cartilaginous matrix; c) substantially removing any antigen derived from said chondrogenic cells a matrix suitable of implantation into a living individual mammal, such as a human being is obtained.

In particular, the invention relates to a method for arthroscopic or endoscopic implantation of homologous or autologous cells into a defect of an animal body, the method comprising a step of I) arthroscopic or endoscopic application of a fluid to a cavity or surface containing the defect, and the steps of ii) application of the cells to the defect substantially simultaneously with a support material, the application being performed at the defect covered by the fluid, iii) mixing of the cells and the supporting material, iv) solidification of the supporting material so that the defect is covered by a mixture of cells and support material without any significant amount of fluid, and v) optionally, removal of the fluid from the cavity or surface by drainage or suction.
