The present invention provides a method for culturing endothelial progenitor cells derived from human umbilical cord blood. According to the present invention, the endothelial progenitor cells derived from human umbilical cord blood cultured zare capable of having the cell adhesive ability and proliferation ratio similar to a cell cultured by a conventional cell culturing method. According to the present invention, collagen derived from umbilical cord and umbilical cord extract can be used as serum replacement and adjurvants for culturing a stem cell and a general animal cell. According to the present invention, a vascular endothelial (precursor) cell, and extract and collagen derived from umbilical cord, which are an autologous substance, are capable of preventing an immune rejection response generated when a cell is transplanted; and minimizing the pollution caused by a pollution source derived from an animal by excluding the use of a protein source derived from an animal. The present invention provides a pharmaceutical composition comprising endothelial progenitor cells derived from human umbilical cord blood for preventing or treating ischemic diseases.

본 발명은 의료용 복합 생체소재에 관한 것이다. 보다 상세하게는, 본 발명은 콜라겐 및 히알루론산 유도체를 포함하는 의료용 복합 생체소재에 대한 것이다. 또한, 상기 생체소재 및 포유류의 탯줄 유래 줄기세포를 이용한 연골세포치료제에 관한 것이다. 이러한, 생체소재는 면역 반응을 야기하지 않고, 내구성이 우수하며, 이러한 생체소재와 줄기세포를 포함한 연골세포치료제는 관절경을 이용한 시술이 가능하여 환자의 고통을 줄여줄 뿐만 아니라 효과적으로 퇴행성 관절염 및 연골 손상을 치료할 수 있다.

PURPOSE: A method for effectively extracting an active ingredient from umbilical cord is provided to use an umbilical cord extract to a cosmetic composition for dressing the skin and improving skin condition. CONSTITUTION: A method for preparing a mammalian umbilical cord extract comprises: a step of cutting umbilical cord; a step of adding the cut umbilical cord to a buffer solution; a step of stirring the mixture at 4-10 deg.C.; and a step of centrifuging the resultant at 4-10 deg. C. and collecting supernatant. The umbilical cord is cut in a length of 0.5-2 cm. The mammalian includes human, pig, horse, cow, mouse, rat, hamster, rabbit, goat, or sheep. The umbilical cord extract contains 800-2,200 pg/ml of FGF-2(fibroblast growth factor-2), 1,000-2,000 pg/ml of G-CSF(Granulocyte colony stimulating factor), 500-1,500 pg/ml of MCP-1(monocyte chemoattractant protein-1), and 300-2,000 pg/ml of GRO[growth regulated oncogene as a main ingredients.

The present invention provides a method for effectively extracting useful components from an umbilical cord. The present invention provides an umbilical cord extract including the useful components. The umbilical cord extract according to the present invention can be used as a serum substitute for cultivating ordinary cells and stem cells derived from an animal. Also, the umbilical cord extract according to the present invention, can be used for a filler for tissue repair, dressing, and improving skin condition. In addition, the present invention relates to a medium composition for isolating and culturing stem cells derived from a tissue, such as an umbilical cord, fatty tissues and the like, and a method for isolating and culturing stem cells derived from the tissue using the same.

PURPOSE: A medical composite biomaterial containing collagen and hyaluronic acid derivatives is provided to ensure durability and to effectively treat degenerative arthritis and cartilage damage. CONSTITUTION: A composition contains collagen and hyaluronic acid or a derivative thereof. The collagen is type I collagen and is derived from human umbilical cord. The hyalruonic acid derivative is prepared by crosslinking hyaluronic acid using BDDE(1,4-butandiol diglycidyl ether). A medical composite biomaterial contains the composition. A chondrocyte therapeutic agent contains the stem cells, collagen, and hyaluronic acid or the derivative. The stem cells are cultured in a container coated with cell adhesion proteins using a medium for culturing the stem cells. The cell adhesion protein is the collagen, gelatin, fibronectin, laminin, or poly-D-lysine. A composition for an injection for treating damaged cartilage contains the chondrocyte therapeutic agent. A method for treating damaged cartilage comprises a step of administering the composition to a patient.

CONSTITUTION: A medium composition for isolating or culturing mammalian umbilical cord-derived stem cells contains an umbilical cord extract without serum. Mammal includes human, pig, horse, cow, mouse, rat, hamster, rabbit, goat, or sheep. A method for preparing the umbilical cord extract comprises: a step of adding cut umbilical cord to a buffer solution and stirring; and a step of collecting supernatant from the mixed solution. A method for preparing the stem cells from the mammalian umbilical cord comprises: a step of culturing a blood-removed umbilical cord tissue in a container which is coated with a cell adhesion protein and contains the medium composition; and a step of treating the tissue with a medium containing stem cell lysis enzyme and isolating the stem cells. The cell adhesion protein includes mammalian umbilical cord-derived collagen, gelatin, fibronectin, laminin, or poly-D-lysine.
