본 발명은 자연수변림의 생태 천이를 반영하여 수변녹지 본연의 기능을 확보하고, 식재 후 자연관리에 의한 관리비용을 최소화할 수 있는 섬진ㆍ영산강유역권 수변녹지의 구축방법에 관한 것으로, 수변녹지 본연의 기능을 확보하고, 식재 후 자연관리에 의한 관리비용을 최소화할 수 있다.
Abstract:
PURPOSE: A method of establishing a riparian green zone around watershed is provided to decide a species composition and the density of planting appropriate for a growth environment, and directly establish an ecological plant assembled with multilayer planting. CONSTITUTION: A riparian green zone is established in the protected lowland placed around Keum river watershed. Four units of pine tree, one unit of elm tree, one unit of snowbell, three units of Sapium japonicum, two units of Lindera erythrocarpa makino, and two units of Carpinus laxiflora Blume are planted are planted per a hundred square meter of a planting area. Two units of pine tree, two units of Carpinus laxiflora Blume, one unit of snowbell, one unit of zelkova, one unit of snowbell, one unit of queritron, and one unit of Acer pseudosieboldianum are planted per a hundred square meter of the planting area. One unit of white oak, one unit of Acer mono Max, two units of snowbell, two units of Lindera erythrocapa makino, two units of styrax obassia, two units of Acer pseudosie boldianum, two units of wild pear tree, and one unit of Sapium japoricum are planted per a hundred square meter of the planting area. Two units of Cornus controversa, one unit of Acer mono Max, one unit of Albizia julibrissin Durazz, two units of snowbell, two units of Lindera erythrocarpa makino, two units of Aphananthe aspera, and two units of Acer pseudosieboldianum are planted per a hundred square meter of the planting area. Five units of pine tree, two unit of Carpinus laxiflora Blume, one unit of oriental oak, three units of snowbell, three units of Acer pseudosieboldianum, three units of styrax obassia, and three units of Lindera erythrocarpa makino are planted per a hundred square meter of the planting area. Three units of pine tree, two units of oriental oak, two units of Lindera erythrocarpa makino, four units of Sapium japonicum, four units of fraxinus sieboldiana, two units of snowbell, two units of Carpinus laxiflora Blume, two units of Quercus acutissima Carruth, and two units of white oak are planted per a hundred square meter of the planting area. 본 발명은 (a) 억새 종자를 캘러스 유도 배지에 치상하여 억새 캘러스를 유도하는 단계; (b) 상기 유도된 억새 캘러스와 외래 유전자가 삽입된 식물 발현 벡터를 포함하는 아그로박테리움 튜머파시엔스 ( Agrobacterium
tumefaciens )를 공동배양 배지에서 공동배양하여 형질전환시키는 단계; (c) 상기 공동배양한 식물세포를 항생제가 첨가된 선발 배지에서 선발하는 단계; (d) 상기 선발된 억새 세포를 재분화 개체를 발근시키기 위한 신장 (elongation) 배지에서 신장시키는 단계; 및 (e) 상기 신장된 세포를 발근 유도배지에서 발근을 유도하는 단계를 포함하는 형질전환 억새 식물체의 제조 방법, 상기 방법에 의해 제조된 형질전환 억새 식물체 및 이의 종자에 관한 것이다.

PURPOSE: A method for producing a transgenic Miscanthus plant is provided to obtain Miscanthus plants with high transformation efficiency using callus. CONSTITUTION: A method for producing a transgenic Miscanthus plant comprises: a step of placing a Miscanthus seed on a callus-inducing medium and inducing Miscanthus callus; a step of co-culturing the induced Miscanthus callus and Agrobacterium tumefaciens containing a foreign gene-inserted plant expression vector; a step of culturing the co-cultured plant cells in a selective medium containing antibiotics; a step of elongating the selected Miscanthus cells in an elongation medium; and a step of inducing rooting of the elongated cells in a medium for inducing rooting.
