Abstract:
A kit for discriminating genetic identification among Cervus species through restriction fragment length polymorphism(RFLP) method is provided to supply accurate Cervus sample and be utilized for managing the quality of the Cervus. The method for discriminating genetic identification among Cervus species comprises the steps of: (a) extracting DNA from a Cervus sample; (b) preparing a reaction solution including the extracted DNA, a sense or an anti-sense primer; (c) amplifying the reaction solution; and (d) performing a RFLP on the amplified DNA to genotype it. The discrimination kit analyzes the genetic variation among Cervus species through the RFLP method including a PCR reagent containing a DNA extract reagent, a sense primer or an anti-sense primer and a restriction enzyme.

A method for discriminating genetic identification of ginseng species is provided to be able to supply accurate ginseng raw material through determination of the ginseng species, thereby being used for maintaining quality of the ginseng. The method comprises the steps of: (a) extracting DNA from ginseng sample; (b) method for preparing a reaction solution of primer consisting of the extracted DNA, sense primer(CCAAGTTGCA AACCCATGGT) and anti-sense primer(TTTGATTTCC TTGGCGCATT CC), VIC-primer(VIC-5'TTGGGTGGAT CTCGT-3-MGB) where TagMan MGB probe is attached at 3-terminal of the VIC primer, and FAM-primer(FAM-5'-TTGGGTGGCT CTCGT-3'-MGB) where TagMan MGB probe is attached at 3-terminal of the FAM-primer; (c) amplifying the reaction solution through PCR; (d) performing a real time PCR on the amplified DNA in combination and then analyzing it by comparing a DNA of Panax ginseng binding to the VIC primer obtained from the step(b) and having A nucleotide with a DNA of Panax quinquefolia binding to the FAM primer obtained from the step(b) and having C nucleotide through gene analysis shown in Fig. 1 using an SDS 7000 software so as to determine the Panax ginseng and the Panax quinquefolia.

A method for discriminating genetic identification of Saposhnikovia divaricata(TURCZ.) SCHISKIN is provided to supply accurate Saposhnikovia divaricata(TURCZ.) SCHISKIN raw material by discriminating the species of the Saposhnikovia divaricata(TURCZ.) SCHISKIN through a discrimination kit and be utilized for maintaining the quality of Saposhnikovia divaricata(TURCZ.) SCHISKIN. The method comprises the steps of: (a) extracting DNA from Saposhnikovia divaricata(TURCZ.) SCHISKIN; (b) preparing a reaction solution of primer consisting of the extracted DNA, sense primer(CTAACACGTC AACAATTTGG) and anti-sense primer(ACGGACGTAC AATTCAGTTT); (c) amplifying the reaction solution through PCR; (d) analyzing the total nucleotide sequence of the amplified DNA; and (e) comparing a gene obtained by performing a pyrosequencing technique on the amplified DNA in combination with sequences: AGGGTTCGCAGACAGG, Saposhnikovia divaricata(TURCZ.) SCHISKIN, and sequence: AGGATTCGCAGACAGG, Peucedanum japonicum Thunberg to discriminate the Saposhnikovia divaricata(TURCZ.) SCHISKIN showing a G peak in the 4th nucleotide and Peucedanum japonicum Thunberg showing an A peak in the 4th nucleotide.
