Abstract:
A promoter and entire genome sequence of heat shock protein isolated from a silk worm is provided to express a excess amount foreign gene and easily select transformant of silk worm. A gene sequence of heat shock protein 70 gene denoted by the sequence 1 comprises gene area and promoter area of heat shock protein 70 isolated silk worm. The Cdna sequence of heat shock protein 70 is denoted by the sequence 2. The amino acid sequence denoted by the sequenc 3 is deduced by cDNA sequence. The maximum promoter activation area of heat shock protein 70 gene comprises 367-1517 sequences.
Abstract:
PURPOSE: A method for preparing recombinant human keratinocyte growth factor using Bombyx mori protein disulfide isomerase gene is provided to improve productivity. CONSTITUTION: A method for preparing recombinant human keratinocyte growth factor comprises: a step of transforming insect cell line with a recombinant expression vector containing the gene; a step of simultaneously or gradually transforming insect cell line with a recombinant vector containing human keratinocyte growth factor gene; a step of selecting transformed cell line which expresses Bombyx mori protein disulfide isomerase and human keratinocyte growth factor; and a step of producing the recombinant human keratinocyte growth factor.

An elongation factor-1 alpha promoter of a silk worm is provided to increase the expression of foreign gene and massively produce recombinant protein. An elongation factor-1 alpha gene sequence of the sequence number 1 comprises gene area and promoter of the elongation factor-1 alpha gene. A cDNA sequence of elongation factor-1 alpha gene is denoted the sequence 2. The amino acid sequence denoted by the sequence number 3 is deduced. The maximum promoter activation area of elongation factor-1 alpha gene comprises 1-1542 sequences.
