Abstract: 본 발명은 국내 사과 재배의 주 품종인 '후지'를, 아그로박테리움(Agrobacterium)을 이용하여 형질 전환하는 기술에 관한 것이다. 보다 상세하게는, 외래 유전자가 도입된 아그로박테리움을 이용해 착색이 증진된 사과 '후지' 품종의 우량계통을 양성하여, 종래 착색에 소요되었던 노동력과 비용의 절감 효과를 가져와 생산의 효율성을 높일 수 있는 형질 전환 기술에 대한 것이다.

Abstract: PURPOSE: A development method of SCAR marker linked to a gene for columnar growth habit in apple is provided, which method allows for discrimination of the columnar growth habit in young plant of apple. CONSTITUTION: The development method of SCAR(sequence characterized amplified region) marker linked to a gene for columnar growth habit in apple comprises the steps of: cross-fertilization of a standard type apple of "Fuji" with a columnar growth type apple of "Tuscan" to obtain a descendant apple F1, and analyzing properties of the descendant apple F1; selecting a marker linked to a gene for columnar growth habit through RAPD(Random amplified polymorphic DNA) analysis using BSA(bovine serum albumin); and sequencing the cloned RAPD marker, wherein the RAPD marker linked to the gene for columnar growth habit is a nucleotide sequence WB82670 of SEQ ID NO:1; and the SCAR marker is a pair of SCAR primers having the nucleotide sequences of SEQ ID NO:2 and SEQ ID NO:3.

Abstract: A method for genetic transformation of 'Fuji' apple using Agrobacterium tumefaciens is provided, thereby improving color development in apples, and reducing labor and costs required for color development of apples, so that production efficiency of apples can be improved. The method for genetic transformation of 'Fuji' apple using Agrobacterium tumefaciens comprises the steps of: (1) culturing apple shoots and rooting the cultured apple shoots in a rooting medium; (2) culturing Agrobacterium tumefaciens containing a foreign gene; (3) inoculating the cultured Agrobacterium tumefaciens to the young leaf cuttings of the apple shoots; (4) culturing the Agrobacterium tumefaciens inoculated young leaf cuttings of the apple shoots in a co-culturing medium; and (5) regenerating the cultured young leaf cuttings of the apple shoots in a regeneration selection medium, wherein the foreign gene is DFR or B-peru.
Abstract translation: 提供了使用根癌农杆菌对“富士”苹果进行遗传转化的方法,从而改善了苹果的发色,降低了苹果发色所需的劳动和成本,从而可以提高苹果的生产效率。 使用根癌农杆菌对“富士”苹果进行遗传转化的方法包括以下步骤:(1)在生根培养基中培养苹果芽,培养苹果芽; (2)培养含有外来基因的根癌土壤杆菌; (3)将培养的根癌土壤杆菌接种到苹果芽的幼叶片中; (4)在共培养培养基中培养根癌土壤杆菌接种苹果芽的幼叶片; 和(5)在再生选择培养基中再生培养的苹果芽幼嫩叶片,其中外源基因是DFR或B-Peru。
