Abstract:
본 발명은 실시간 멀티플렉스 중합효소연쇄반응을 이용하여 HLA-A, B, DR 대립유전자를 동시에 검사할 수 있는 방법 및 키트에 관한 것으로 시료에서 추출한 DNA를 대상으로 HLA 대립유전자에 특이적으로 결합할 수 있는 시발체들과 이들 시발체에 의하여 증폭된 증폭산물을 탐지할 수 있는 형광탐침자들을 이용하여 나타나는 형광값을 가지고 HLA 대립유전자의 형별을 구분하는 방법 및 HLA 대립유전자의 형별을 구분하기 위한 키트에 관한 것이다.
Abstract:
PURPOSE: A selection kit for HLA allele is provided to distinguish HLA allele through a real-time polymerase chain reaction without electrophoresis by using flurogenic probe and amplifying the gene in multiplex. CONSTITUTION: A selection kit for HLA allele comprises the following: a primer set capable of selective amplification by real-time PCR about the HLA allele of more than two genes selected from a group comprising HLA-A and HLA-B; and a probe set capable of probing the product amplified by the real-time PCR. A selecting method of HLA allele comprises the following steps: A real-time polymerase chain reaction (PCR) by using the kit to the refined DNA; and a selection of HLA allele by reading the result of PCR.
Abstract:
The present invention relates to a method for selectively amplifying a certain DNA fragment where a short fragment is composed of a reciprocal connection of G and C, and the front of the fragment is processed with phosphorothioate; and the short DNA fragment enables a DNA polymerase with thermal stability not to be amplified autonomously at low temperatures but to be melted at high temperatures, leading to activity recovery.
