Abstract:
본 발명은 인슐린신호경로를 조절하는 miRNA와 그 표적 유전자의 작용을 제어하는 물질을 스크리닝 하는 방법에 관한 것으로, 구체적으로, 세포 성장을 촉진하는 miR-8 또는 miR-200 miRNA의 표적 유전자인 USH 또는 FOG2의 작용을 제어하는 물질을 스크리닝 하는 방법에 관한 것이다. 본 발명자들은 초파리 지방체 세포에서 u-shaped(ush )를 표적함으로써 체구를 조절하는 보존된 miRNA인 miR-8을 발굴하였다. 또한, 초파리 miR-8 miRNA의 인간 상동 유전자인 miR-200의 표적 유전자가 FOG2임을 확인하였다. 초파리의 miR-8 및 USH가 포유동물에서도 보존되어 있으며, USH의 인간 상동 유전자인 FOG2가 PI3K의 조절 서브유니트와 직접 결합하여 기능함을 발견하였다. 인간 암세포주에서 miR-200의 발현을 억제하거나 FOG2를 발현시켰을 때 세포 성장을 촉진하는 PI3K 활성이 감소되는 것을 확인하였으므로, miR-200 및 FOG2를 인슐린신호경로 조절물질의 스크리닝에 유용하게 사용할 수 있다.
Abstract:
The present invention relates to an amphiphilic peptide capable of promoting target miRNA production and a method for regulating the production of target miRNA using the same. In detail, the amphiphilic peptide of the present invention binds strongly and specifically to hairpin-shaped target miRNA. The specific binding affinity induces the Dicer enzyme activity, therefore specifically increase the production of target miRNA. The present invention can be effectively used for regulating the amount of target miRNA produced in vivo, for the study of miRNA functions and for producing therapeutic drug for target miRNA related disease.
Abstract:
PURPOSE: A method for measuring microRNA activity is provided to detect uridinylation and screen a test sample compound which affects uridinylation when miRNA is generated by detecting uridinylation. CONSTITUTION: A method for screening uridinylation controlling compound comprises: a step of transducing an expression vector which expresses Lin28 protein in a test cell line; a step of treating a test sample compound to the transformed test cell line; and a step of screening uridinylated miRNA by grinding the treated test cell line.

PURPOSE: A PI3K activation controlling agent containing a fifth zinc finger domain of FOG2 is provided to induce cancer cell apoptosis and to prevent and treat PI3K-associated diseases. CONSTITUTION: A polypeptide contains a fifth zinc finger domain of FOG2(Friend of GATA 2). The polypeptide has 20-500 amino acids. The polypeptide contains 547-570th amino acid sequence at N-terminal of FOG2 amino acid sequence of sequence number 1. An agent for anticancer or anticancer aid contains the polypeptide; a nucleic acid molecule encoding the polypeptide; or vector containing a nucleic acid molecule. A composition for preventing and treating metabolic disease contains the polypeptide, the nucleic acid encoding the polypeptide, or the vector.

PURPOSE: A method for screening a material regulating the function of USH or FOG2 which is miR-8 or miR-200 miRNA is provided. CONSTITUTION: A method for screening a material regulating insulin signal transduction comprises: a step of treating a test compound to a cell line which expresses miR-200 family miRNA or miR-8 miRNA; a step of measuring expression level or activity of FOG2 or USH protein in the cells; and a step of screening a test compound which changes the expression or activation of FOG2 or USH protein.
