Abstract: PURPOSE: Recombinant E. coli with reconstructed sucrose metabolic pathways is provided to be used in a microorganism-based industry by transforming E.coli which produces a secondary metabolite using only sucrose as a carbides source. CONSTITUTION: Sucrose-using E.coli is a reconstructed PTS(phosphoenolpyruvate (PEP)-dependent phosphotransferase transport system) system or non-PTS system. The PTS system is prepared by transduction of sucrose phosphotransferase, sucrose hydrolase, and fructokinase. The non-PTS system is prepared by transduction of sucrose permease, sucrose hydrolase or phophorylase, and fructokinase. The PTS-syste-introduced E.coli is prepared by transformation with pAC-AAK plasmid which expresses scrA, sacA, and sack genes. The gene is derived from Staphylococcus xylosus and Lactobacillus plantarum.
