Abstract: PURPOSE: A composition containing death receptor 5(DR5) is provided to induce cancer cell apoptosis or TRAIL-sensitivity and to prevent or treat cancer. CONSTITUTION: An anti-DR5 antibody which specifically binds to DR5 comprises an amino acid of sequence number 7 or 39. The anti-DR5 antibody contains heavy chain variable region of amino acid sequence numbers 1, 2, and 3 and light chain variable region of amino acid sequence of sequence numbers 4, 5, and 6. The light chain variable region has an amino acid of sequence number 27 or 37. The heavy chain variable region has an amino acid sequence of sequence number 28 or 38.

Abstract: An antibody is provided to be effectively used for preventing or treating various cancers through inducing autophagic cell death of TRAIL-sensitive cancer cells as well as TRAIL-resistant cancer cells by specific binding to death receptor 5(DR5). An antibody specifically binding to DR5 is selected from the group consisting of an antibody comprising a heavy chain variable region(VH) having the amino acid sequences of SEQ ID NOs: 1 to 3 at complementary determining regions(CDRs) and a light chain variable region(VL) having the amino acid sequences of SEQ ID NOs: 4 to 6 at CDRs; and an antibody comprising a VH having the amino acid sequences of SEQ ID NOs: 7 to 9 at CDRs and a VL having the amino acid sequences of SEQ ID NOs: 10 to 12 at CDRs. A composition for preventing or treating a cancer comprises the antibody as an active ingredient, wherein the antibody is a whole antibody selected from the group consisting of IgG, IgM, IgA, IgD and IgE or an antibody fragment selected from the group consisting of scFv, (scFv)2, Fab, Fab', and F(ab')2 and the cancer is selected from the group consisting of breast cancer, colon cancer, brain tumor, glioma, ovarian cancer, endometrial cancer, bone sarcoma, cervix cancer, prostatic cancer, lung cancer, Synovial cancer, pancreatic cancer and sarcoma.

Abstract: 본 발명은 세포사멸 수용체 5 (death receptor 5; DR5)에 특이적으로 결합하는 항체 및 이를 포함하는 암 예방 또는 치료용 조성물에 관한 것으로, 상보성 결정 영역에 서열번호 1 내지 3의 아미노산 서열을 갖는 중쇄 가변영역 및 서열번호 4 내지 6의 아미노산 서열을 갖는 경쇄 가변영역을 포함하는 항체; 및 상보성 결정 영역에 서열번호 7 내지 9의 아미노산 서열을 갖는 중쇄 가변영역 및 서열번호 10 내지 12의 아미노산 서열을 갖는 경쇄 가변영역을 포함하는 항체로 이루어지는 군으로부터 선택되고, DR5에 특이적으로 결합하는 본 발명의 항체는 TRAIL-민감성 암세포뿐만 아니라 TRAIL-저항성 암세포에서도 DR5를 통한 자식작용에 의한 세포사멸 (autophagic cell death)을 유도하므로, 다양한 암의 예방 또는 치료에 사용될 수 있다.

Abstract: 본 발명은 세포사멸 수용체 5(death receptor 5; DR5)에 특이적으로 결합하여 다양한 암세포를 효과적으로 사멸하는 친화도와 안정성이 향상된 항 ＤＲ5 항체, 및 이를 포함하는 암 예방 또는 치료용 조성물에 관한 것이다. 본 발명에 따른 항 ＤＲ5 항체는 기존의 항 ＤＲ5 항체인 HW1의 중쇄가변영역 내의 VH-CDR2, VH-CDR3의 아미노산 잔기와 경쇄가변영역 내의 VL-CDR3의 아미노산 잔기를 돌연변이화하고, 중쇄가변영역 내의 구조(framework)를 VH6 아류형에서 VH3 아류형으로 치환하고 경쇄가변영역 내의 구조를 Vk1 또는 Vk3의 구조 부분으로 치환함으로써, 기존의 항 ＤＲ5 항체인 HW1보다 친화도와 안정성을 향상시킨 것을 특징으로 한다. 또한, 본 발명에 따른 항 ＤＲ5 항체는 DR5를 발현하는 TRAIL-민감성 암세포 또는 DR5를 발현하는 TRAIL-저항성 암세포에 대해 자식작용(autophagy)에 의하여 세포사멸을 효과적으로 유도함으로써, ＤＲ5 발현에 의해 야기되는 암의 예방 또는 치료에 유용하게 사용될 수 있다.
