Abstract:
A conjugate of a hapten compound of metamifop and a protein is provided to be used as an antibody for rapidly and quantitatively detecting the metamifop. A conjugate is characterized in that a hapten compound of metamifop is bound to a protein by an amide bond. The hapten compound of the metamifop is represented by the formula(1), wherein each R1, R2, R3, and R4 is independently H, carboxyl, hydroxy, C1-10 alkyl, halogen, cyano, amino, thiol, aldehyde, nitro, C2-10 carboxyalkyl or C1-10 hydroxyalkyl; each R5 and R6 is independently H, carboxyl, hydroxy, C1-10 alkyl, halogen, cyano, amino, thiol, aldehyde, nitro, C1-10 carboxyalkyl, C1-10 hydroxyalkyl or a group represented by the structural formula(1-a); R7 is H, C2-10 carboxyalkyl or C1-10 alkyl; and each R8, R9, R10, R11 and R12 is independently H, carboxyl, hydroxy, C1-10 alkyl, halogen, amino, thiol, aldehyde, C2-10 carboxyalkyl or C1-10 hydroxyalkyl, provided that at least one of the R1 to R12 is at least one of carboxyl and amino. The protein is bovine serum albumine(BSA), human serum albumine(HSA), fibrinogen, ovalbumin, thyroglobulin or keyhole limpet hemocyanin(KLH). An antibody is produced by being reacted to the conjugate and is specifically bound to the metamifop. A method for producing the antibody comprises the steps of: (a) immunizing mammals with the conjugate; and (b) selecting the antibody specific to the metamifop from the mammals.

A hapten-protein complex and an antibody used to an enzyme-linked immunosorbent analysis of an insecticide bistrifluron residue are provided to detect quickly and quantitively the insecticide bistrifluron using the antibody to a composite in which a specific hapten compound of bistrifluron and a protein are combined. A composite is manufactured by combining a hapten compound of an insecticide bistrifluron and a protein using an amino bond. The hapten compound of an insecticide bistrifluron includes a carboxyl group or an amino radical and a compound of haptens I, II, III, IV or V. The protein is a bovine serum albumin(BSA), a human serum albumin(HSA), a fibrinogen, an ovalbumin, a thyroglobulin or a keyhole limpet haemocyanin(KLH).

An enzyme immunology analysis method for detecting insecticide bistrifluron residue is provided to detect quickly and quantitively the insecticide bistrifluron using the antibody to a composite in which a specific hapten compound of bistrifluron and a protein are combined. A method for detecting a bistrifluron in a sample comprises steps of: contacting an antibody generated from a composite, a sample including insecticide bistrifluron and the composite a composite in which a hapten compound of a bistrifluron fixed to a solid support and a protein are combined using an amino bond; and detecting a bond of the antibody to the bistrifluron fixed to the solid support.

A method for detecting metamifop in a sample comprises the steps of: (a) contacting an antibody generated from a complex in which hapten compound of metamifop and protein are bound by an amide bond with the sample including metamifop and a complex in which a hapten compound of metamifop fixed on a solid support and protein are bound by an amide bond; and (b) detecting the binding of the antibody regarding the metamifop using an enzyme labeled secondary antibody, wherein the hapten compound of the metamifop includes a carboxyl or an amino group, the protein is bovine serum albumine(BSA), human serum albumine(HSA), fibrinogen, ovalbumin, thyroglobulin or keyhole limpet hemocyanin(KLH).

본 발명은 살충제 비스트리플루론(bistrifluron)을 포함하는 시료와 비스트리플루론의 합텐 화합물과 단백질이 아미드 결합에 의해 결합된 복합체로부터 생성된 항체 및 고체 지지체에 고정된 비스트리플루론의 합텐 화합물과 단백질이 아미드 결합에 의해 결합된 복합체를 접촉시키는 단계 및 상기 고체 지지체에 고정된 비스트리플루론에 대한 상기 항체의 결합을 검출하는 단계를 포함하는, 시료 중의 비스트리플루론을 검출하는 방법에 관한 것이다. 본 발명에 따르면, 비스트리플루론의 특정 합텐 화합물과 단백질이 결합된 복합체에 대한 항체를 이용함으로써 살충제 비스트리플루론을 신속하고 정량적으로 검출할 수 있다.
