Abstract:
A method for preparing a phenylpropanoid amid compound is provided to mass-product the phenylpropanoid amid compound, which has various pharmacological activities such as anti-bacterial, anti-oxidative, and bone formation promoting activity, such as coumaroylserotonin, feruloylserotonin, and caffeoylserotonin conveniently and economically using methanol as an elicitor. A biosynthesis inducer of phenylpropanoid amide compounds in plants or an expression inducer of serotonin hydroxycinnamoyl transferase(SHT) or tryptophane decarboxylase(TDC) in the plants comprises methanol as an elicitor. To biosynthesize phenylpropanoid amide compounds in a plant, the plant is treated with methanol. To induce expression of SHT or TDC in a plant, the plant is treated with methanol. Further, a concentration of the used methanol is 0.5 to 1.0%.

본 발명은 P450 효소의 촉매활성을 증대시키는 CPR 유전자를 포함하는 재조합 벡터, 상기 재조합 벡터 및 P450 유전자를 포함하는 재조합 벡터에 의하여 형질전환된 세균 및 이를 이용한 P450 촉매반응 화합물의 제조방법에 관한 것이다. 본 발명에 따른 재조합 벡터는, P450 효소의 촉매활성을 증대시키는, 서열번호 12의 염기서열로 구성되는 벼 유래의 NADPH-시토크롬 P450 리덕타제2(NADPH-cytochrome P450 reductase2) 유전자를 포함하는 재조합 벡터를 포함한다. 또한 본 발명에 따른 형질전환 세균은, 서열번호 12의 염기서열로 구성되는 벼 유래의 NADPH-시토크롬 P450 리덕타제2(NADPH-cytochrome P450 reductase2) 유전자를 포함하는 재조합 벡터와; 트립타민 5-하이드록시다아제(tryptamine 5-hydroxylase) 유전자를 포함하는 재조합 벡터 또는 시나메이트 4-하이드록시다아제(cinnamate 4-hydroxylase) 유전자를 포함하는 재조합 벡터에 의하여 형질전환된 세균을 포함한다. 상기 형질전환된 세균을 통하여 세로토닌 또는 쿠마르산을 안정적으로 다량 생산할 수 있다.

The present invention relates to a recombinant vector comprising CPR gene which increases catalytic activity of P450 enzyme, bacteria transformed by the recombinant vector and a recombinant vector comprising P450 gene, and a method for producing a P450 catalysis compound using the same. The recombinant vector according to the present invention comprises a recombinant vector which increases the catalytic activity of P450 enzyme and has a rice-derived NADPH-cytochrome P450 reductase 2 gene consisting of base sequences of sequence number 12. The bacteria according to the present invention comprises: the recombinant vector having the rice-derived NADPH-cytochrome P450 reductase 2 gene consisting of base sequences of sequence number 12; and a recombinant vector comprising tryptamine 5-hydroxylase gene or a recombinant vector comprising cinnamate 4-hydroxylase gene. Serotonin or coumaric acid is stably produced in large amounts using the transformed bacteria.

A transformed bacteria by a recombinant vector having a 4CL gene and SHT gene is provided to massively produce phenylpropanoid amide-based compound and reduce production cost. A recombinant vector comprises a serotonin N-hydroxycinnamoyl transferase (SHT) gene, 4-coumarate-CoA ligase (4CL) gene, and a promoter. A method for producing a phenylpropanoid amide-based compound comprises: a step of injecting 0.5mM~5mM of each phenol substrate and amine substrate in a transformed bacteria and culturing at 28~40°C for two to four hours; and a step of isolating and purifying the produced phenylpropanoid amide-based compound from the bacteria.
