Abstract:
PURPOSE: A recombinant vector for a development/secretion of a foreign protein is provided to improve the secretion efficiency and to confirm the development of the foreign protein by combining and epitope. CONSTITUTION: A recombinant vector for a development/secretion of a foreign protein contains a signal peptide, a protein development verifier epitope and an insertion site for the foreign protein. The signal peptide is either an OmpW signal peptide, a StfA signal peptide or a AgfA signal peptide. The protein development verifier epitope is either a His epitope, an RGS-His epitope, a T7 epitope or a V5 epitope. The insertion site for the foreign protein comprises an EcoRI, BamHI, SalI and HindIII cutting region. The producing method of the foreign protein comprises the following steps: culturing a transformant obtained by transforming a host cell using the recombinant vector; and confirming the foreign protein at a western blot.

PURPOSE: A live vaccine composition containing bacteria incapable of surviving at the environment is provided to easily produce and store the vaccine, and to safely and effectively prevent diseases when injecting to domestic animals. CONSTITUTION: A live vaccine composition containing auxotrophic bacteria which is lack of essential genes necessary for the outgrowth of a cell, and need to be supplied with the essential genes from a auxotrophy recombinant vector. The auxotrophic bacteria are either the Salmonella typhi, the S. typhimurium, the S. enteritidis, the S. galinarum, the S. dublin, the S. Cholerae-suis or the S. pullorum. The essential genes necessary for the outgrowth of the cell is aspartate kinase, aspartate semialdehyde dehydrogenase, dihydrodipicolinate synthase, dihydrodipicolinate reductase, tetrahydrodipicolinate reductase, glutamate:succinyl-diaminopimelate aminotransferase, or succinyldiaminopimelate desuccinylase.

An attenuated Salmonella live vaccine is provided to be easily and economically produced and stored and prevent Salmonella infection safely and efficiently when administered to a domestic animal. A method for preparing a Salmonella live vaccine comprises a step of deleting a lon or a cpxR gene from a chromosome DNA of a Salmonella strain such as Salmonella typhimurium, S. typhi, S. paratyphi, S. sendai, S. gallinarium and S. enteritidis. A vector includes a DNA where a DNA fragment linked to a 5'-terminal of the lon gene derived from the Salmonella strain and a DNA fragment linked to a 3'-terminal of the lon gene are bonded. An attenuated Salmonella vaccine composition comprises a Salmonella mutant where the lon gene and the cpxR gene are deleted from the chromosome DNA of the Salmonella and a pharmaceutically acceptable carrier.
