    Abstract: PURPOSE: A method for isolating hypnospores of Plasmodiophora brassicae and a method for proliferating using the same are provided to enable molecular biological study. CONSTITUTION: A method for isolating hypnospores from infected plants or soil comprises: a step of pretreating infected plants or soil to washed Plasmodiophora brassicae bacteria; a step of removing impurities from the plants or soil using a fabric made of cotton, silk, rayon, or synthetic fiber; a step of isolating prepared suspension with a cellulose filter; and a step of centrifuging the isolated suspension and centrifuging to prepare a pellet. The Plasmodiophora brassicae bacteria is Plasmodiophora brassicae.

    Abstract: PURPOSE: A primer which specifically binds to clubroot and a method for diagnosing clubroot of Brassicaceae crops using the same are provided. CONSTITUTION: A method for diagnosing clubroot of Brassicaceae crops comprises: a step of performing PCR of DNA isolated from a spore of Brassicaceae crops from crops and soil under the presence of a primer set which specifically amplifies ITS(interual transcribed spacer) gene; and a step of analyzing gene amplification to determine club root of Brassicaceae crops. The primer set contains a forward primer of sequence number 1, 3, 5, or 7 and a reverse primer of sequence number 2, 4, 6, or 8.
