Abstract:
PURPOSE: A double-specific gene expression system for a neuroglial tissue and an ischemic tissue is provided to simultaneously recognize the tissues and to effectively treat or prevent ischemic cerebral diseases. CONSTITUTION: A double-specific recombinant vector for a neuroglial tissue and an ischemic tissue contains etythropoietin enhancer, nestin intron, promoter, and therapeutic agent for ischemic cerebral diseases. The enhancer contains a base of sequence number 1. The netin intron contains a base of sequence number 2. The promoter has an ischemic tissue-selective promoter sequence. The recombinant vector is a viral vector. A pharmaceutical composition for treating ischemic cerebral diseases contains the recombinant vector as an active ingredient.

PURPOSE: An effective expression system of low oxygen-selective therapeutic gene using tyrosine hydroxylase 3' UTR is provided to treat ischemic diseases. CONSTITUTION: A recombinant vector for ischemia-specific expression contains a promoter, ischemia therapeutic gene, and tyrosine hydroxylase 3' UTR(untranslated region) sequence fused at the gene terminal. The ischemia therapeutic gene is a vascularization factor, such as basic fibroblast growth factor(bFGF), vascular endothelial growth factor(VEGF), plateletderived growth factor(PDGF), hepatocyte growth factor(HGF), heme oxygenase 1(HO-1), insulin growth factor(IGF-I or II), heat shock protein 27(HSP27), or tymidine kinase. A pharmaceutical composition for treating ischemia contains the recombinant vector as an active ingredient.

CONSTITUTION: A recombinant expression vector contains a ischemia therapeutic gene and an oxygen dependent degradation domain(ODD domain) of ATF-4(activating transcription factor-4). The ischemia therapeutic gene is a vascularization factor. The ATF-4 ODDD sequence is derived from human and contains a base sequence of sequence number 1.
