Abstract: PURPOSE: To rapidly generate a highly reproducible result by providing negative and positive reference areas to the reaction surface area of a solid phase assay apparatus and allowing both areas to show the effectiveness of a test result in the presence of a label forming a display signal. CONSTITUTION: In an analyser 10, a matrix 12 is arranged within a carrier 14 and built-in negative and positive areas 30, 32 and an analyzing substance bonding area 34 for bonding an analyzing substance are provided on the matrix 12. The reference area 32 is formed in a form of a -code and the bonding area 34 is formed in a rod shape oriented vertically to the reference area 32. With respect to a positive test result obtained from the correct use of the analyser 10, the reference area 32 and the bonding area 34 act mutually to generate reaction detectable in a form of a +code to show the positive test result. With respect to a negative test result obtained from the correct use of the analyser 10, reaction is generated in a form of a -code only in the reference area 32 to show the negative test result. When bonding assay is not correctly executed, detectable reaction is not obtained in the reference area 32 or the bonding area 34.

Abstract: PURPOSE: To measure a biological fluid with high sensitivity by providing a porous matrix of fiber and solid particles, wherein the particles have a substance reacting on an analyte in a sample to the surface thereof and held fixedly in the matrix. CONSTITUTION: A material for an analytical unit 10 comprises a porous fiber matrix. The fibrous material has a mean diameter of about 0.1-10μm and has a plurality of spherical solid particles, being held fixedly on the fiber. The negative control region 30 of the matrix 12 does not contain a substance holding an enzyme label, and a signal reactive material within a control region 32 is provided with a substance to be bonded with an enzyme label and other signal reactive materials. In an analyte bonding region 32, microparticles in an area 34 of the matrix 12 is coated with a substance, e.g. an antigen or antibody, to be bonded with the analyte. Positive test results obtained through the correct use of the unit 10 causes '+' reaction both in the control region 32 and the analyte bonding region 34, while negative test results causes '-'reaction in the control region 32, thus providing valid test results. When the reagent does not function correctly, no reaction takes place in the control region 32 or the analyte bonding region 34, and invalid test results are provided.

Abstract: PURPOSE: To obtain accurate results with high sensitivity and reproducibility through simple operation by providing a reaction matrix with a specified positive/negative control region and an analyte bonding region. CONSTITUTION: The solid phase analyzer 10 comprises a reaction matrix 12 provided in a carrier 14, a barrier substance 18, a suction means 20, and a filter means 22. The matrix 12 is composed of micro solid particles fixed onto a fibrous substance having a diameter of 0.1-10 micron. The matrix 10 is provided with a negative control region 30, a positive control region 32, and an analyte bonding region 34. The region 30 does not contain any substance for holding an enzyme label or other sign responsive substance but the region 32 contains that substance and micro particles in the region 34 are applied with a substance to be bonded with the analyte, e.g. an antigen or an antibody. A sample liquid containing an antigen or antibody is applied to the matric 12 through the filter 22 and the response is detected by sandwich immunoassay under presence of an enzyme linked antibody.

Abstract: A novel material and device useful in solid-phase binding assays to determine the presence or amount of an analyte in a test sample, particularly antigens or antibodies, is disclosed. The material comprises a porous matrix of fibers and a plurality of substantially spherical, solid particles having an average diameter of from about 0.1 to about 5 microns. The particles are retained and immobilized upon the fibers of the matrix. Preferably, the particles have on their surfaces a substance capable of reaction with the analyte in the sample, and the average diameter of the particles is less than the average pore size of the matrix. The device, in a preferred embodiment, comprises a substantially planar layer of the described material.
