Date Jul. 31, 1986.The invention relates to a procedure of obtaining a lipid mixture rich in the content of gamma -linolenic acid or glycerides thereof from cultured fungus bodies of a filamentous Mortierella fungus. The invention proposes, on one hand, to fractionally extract the lipid mixture from the fungus bodies under mechanical disintegration in two steps first with an alcohol containing water to give a fraction rich in the polar lipids and then with a hydrocarbon solvent, e.g. hexane, to give a fraction lean in the polar lipids. The invention proposes, on the other hand, to enrich a lipid mixture relative to gamma -linolenic acid by fractionally crystallizing a part of a starting lipid mixture in a solution of a specified solvent so that the uncrystallized fraction in the mother liquor contains a lipid mixture in which the content of gamma -linolenic acid is remarkably increased in comparison with the starting lipid mixture.

    Abstract: A very efficient microbiological method is proposed for the preparation of gamma -linolenic acid or a lipid rich in the content of gamma -linolenic acid, one of the essential fatty acids in the diet of mammals, e.g. human. The method comprises culturing a fungus of Mortierella genus including isabellina, vinacea, ramanniana, ramanniana var. angulispora and nana in a culture medium containing a carbohydrate, e.g. glucose, as the carbon source in an unusually high concentration of 60 to 400 g/liter to grow a fungal body containing a large amount of a lipid rich in the content of gamma -linolenic acid. A further improvement in the efficiency of culture and the content of the desired fatty acid in the fungal body is obtained by the addition of acetic acid or an alkali acetate to the culture medium.

    Abstract: A METHOD FOR THE PREPARATION OF A FUNGAL BODY AND A LIPID RICH IN .gamma.-LINOLENIC ACID THEREFROM A very efficient microbiological method is proposed for the preparation of .gamma.-linolenic acid or a lipid rich in the content of .gamma.-linolenic acid as one of the essential fatty acids in the diet of mammals, e.g.human.     Abstract: PURPOSE:To provide the subject factor extracted and separated from intercellular substance of spleen tissue and having activity to promote the proliferation of hepatocyte in vitro. CONSTITUTION:Fresh spleen is extracted from a slaughtered animal such as rat, cattle or pig, placed on a mesh of e.g., stainless steel and crushed to loosen the cell cluster. The cells are washed with a proper amount of a physiological salt solution such as physiological salt solution buffered with phosphoric acid to collect the intercellular substance in a container placed under the mesh. The obtained intercellular substance is centrifuged and a soluble fraction containing the hepatocyte-proliferation promoting factor is obtained as the supernatant liquid. The proliferation factor has the following characteristics. (i) Molecular weight is 40,000 and 150,000 or more. (ii) It is not adsorbed to a heparin column. (iii) The hepatocyte-proliferation promoting activity is inactivated by the heat-treatment at 100 deg.C for 2min. (iv) The hepatocyte-proliferation promoting activity is not inactivated in the presence of trypsin (treated at 34 deg.C for 2hr at a concentration of 0.01%). (v) It has an activity to effect the proliferation of hepatocyte in vitro.

    Abstract: PURPOSE:To obtain the titled composition, containing a gamma-linolenic acid- containing fat and oil extracted from fungal cells of the genus Mortierella, annually producible and available at a low cost and having excellent skin touch and simultaneously producing various effects originating from the gamma-linolenic acid. CONSTITUTION:A cosmetic or dermal composition obtained by containing a gamma-linolenic acid-containing fat and oil extracted from cells of fungi which are molds belonging to the genus Mortierella. This composition can be applied in the form of a water-in-oil type or oil-in-water type emulsion system, e.g. milky lotion, lotion, shampoo or shaving foam, or more concentrated emulsion system, e.g. facial pack. In this case, the fat and oil of the fungal cells can be contained in an amount of 1-20wt.%. The above-mentioned composition may consists essentially of an oily phase or aqueous phase and is applicable in the form of a solid, e.g. makeup product. In this case the composition can be contained in amount of about 1-30wt%.

    Abstract: PURPOSE:To produce a neutral lipid and polar lipid in high productivity, by culturing a microorganism belonging to Mortierella genus. CONSTITUTION:A microbial strain selected from Mortierella isabellina, M. vinacea, M. ramanniana vas. anglispora or M. nana, e.g. Mortierella isabellina IFO 7884 is inoculated in a medium containing a carbohydrate as a carbon source and cultured under aerobic condition at 4.0-6.0pH for about 2-15 days. The objective neutral lipid and polar lipid can be separated from the cultured microbial cell.

    Abstract: PURPOSE:To obtain refined glyceride oil effectively removing a gum such as phospholipid, diglyceride, free fatty acid, etc., by extracting crude glyceride oil composition from microorganism, diluting the composition with an organic solvent and contacting with a semipermeable membrane under pressure. CONSTITUTION:A crude glyceride oil composition extracted from microorganism, e.g. from the microbial cell of Mortierella genus is diluted with an organic solvent and the solution is made to contact with one surface of a semipermeable membrane under pressure. The liquid permeated to the other surface of the membrane is collected and the organic solvent is removed therefrom to obtain a glyceride oil having decreased amount of foreign materials. The semipermeable membrane is preferably made of a polyimide polymer, e.g. a polymer having the recurring unit of formula I [R is bivalent aromatic group including the group of formula II [X is -CH2-, -O-, etc.) and formula III (Y is -CH3, etc.) or unit of formula IV (R and R are vibalent aromatic group), especially preferably a polyimide composed of the recurring units of formula V and formula VI.
