    Abstract: Techniques for producing cloned DNA sequences are provided which sequences are complementary to DNA occurring in one selected region of one chromosome of a multi-chromosomal genome, such as the human genome. Such cloned DNA sequences can be labeled and formed into probes by conventional procedures, there are provided methods for making probe compositions which comprise mixed DNA segments derived from such a DNA sequence. An improved DNA sequence transamination procedure is provided utilizing trifluoroacetate chaotrope anions. With high concentrations of low complexity DNA, high levels of transamination are thereby achieved. These segments are covalently bound to fluorophore groups through linking groups that are transaminated preferably chaotropically into the segments.

    Abstract: Sont décrites des techniques pour produire des séquences d'ADN cloné qui sont complémentaires de l'ADN se trouvant dans une région choisie d'un chromosome d'un génome multichromosomique tel que le génome humain. Ces séquences d'ADN cloné peuvent être marquées et transformées en sondes par des procédés classiques; sont décrites des méthodes pour fabriquer des compositions de sondes qui renferment des segments d'ADN mixte dérivés d'une telle séquence d'ADN. Un procédé perfectionné de transamination de séquences d'ADN fait appel à des anions chaotropes de trifluoracétate. On obtient ainsi des niveaux élevés de transamination avec des concentrations élevées d'ADN de faible complexité. Ces segments sont liés par covalence à des groupes fluorophores par l'intermédiaire de groupes de liaison qui sont transaminés de préférence par voie chaotropique dans les segments.
