Abstract:
Methods for identifying inducers and inhibitors of programmed cell death in a cell-free system are described. The methods exploit the finding that programmed cell death is accompanied by shutdown of cellular protein synthesis and by phosphorylation of eIF-2α and that the dephosphorylation of eIF-2α prevents the shutdown of protein synthesis.

Un método para identificar inhibidores de la muerte celular programada en un sistema libre de células, comprendiendo el método las etapas de: (a) preparar un sistema libre de células que comprende eIF-2 fosforilado y proteína fosfatasa 1 (PP1); (b) introducir en el sistema libre de células de la etapa (a) un inhibidor candidato de la muerte celular programada, y dejar incubar a la mezcla resultante; y (c) medir el nivel de fosforilación de eIF-2 resultante de la etapa (b); en el que un inhibidor de la muerte celular programada disminuye o evita el aumento del nivel de fosforilación de eIF-2 que acompaña a la muerte celular programada, cuando se compara con un control.
