    Abstract: A solid-phase nucleotide synthesis intermediate useful for chemical synthesis of oligodeoxyribonucleotides and oligoribonucleotides comprises: (1) a particulate support comprising a porous polymer whose backbone comprises optionally substituted acrylate or methacrylate moieties; (2) a nucleoside; and (3) a linker having a first and a second end, the first end being covalently attached to the particulate support and the second end being covalently attached to the nucleoside, the linker spacing the nucleoside at least 3 atoms away from the polymer. The linker can comprise at least one optionally substituted aliphatic diamine. Alternatively, the linker can comprise a polyethylene glycol moiety. Preferably, the porous polymer is a methacrylate-vinylidene polymer. The solid-phase support thus produced can be used for oligodeoxyribonucleotide synthesis by either the phosphite-triester or the phosphotriester processes.

    Abstract: Universal solid support oligonucleotide synthesis reagents, oligonucleotide synthesis processes, and reagents for cleaving oligonucleotides from solid supports are disclosed. Oligonucleotide synthesis reagents have the following general formula: SS - R6 - O - R3 wherein SS is a solid support; R6 is (a), (b) or (c), where R5 is hydrogen or alkyl and R4 is a phosphate protecting group; and R3 is a ring moiety having vicinal groups -XR1 and -YR2 wherein each of X and Y is independently selected from the group consisting of O, S and NH and one of R?1 and R2¿ is a blocking moiety and the other is hydrogen or a hydroxy protecting group. Oligonucleotide cleaving reagents include methylamine and/or ammonium hydroxide and trimethylamine.

    Abstract: Novel chemiluminescent compounds are suitable for use in labeling biological molecules for assays such as immunoassays. These chemiluminescent labels are characterized by the incorporation of stable leaving groups. One class of these chemiluminescent labels comprises salts in which the leaving group contains a carboxyl carbon atom or its isoelectronic equivalent and a five-membered ring, including at least one heteroatom. The heteroatom is preferably oxygen or sulfur. This leaving group is linked to a positively charged moiety capable of producing light by chemiluminescence, which can be an acridinium, phenanthridinium, quinolinium, or benzacridinium moiety. Both the leaving group and the positively charged moiety can be substituted, such as with reactive substituents for covalently linking the label to a biological molecule. Substituents on the five-membered ring of the leaving group can form an additional ring. An additional class of these chemiluminescent labels comprises a chemical group that can produce light by chemiluminescence covalently linked to a leaving group comprising a moiety containing a sulfur, phosphorus, or carbon atom double-bonded to a more electronegative atom or atoms.
