    Abstract: PROBLEM TO BE SOLVED: To provide a two-dimensional electrophoresis apparatus which supplies separability far higher than that obtained by a one-dimensional separation as a useful technology for separating a complex protein mixture. SOLUTION: A strip gel and a slab gel which may be water-expandable are combined on a common support for two-dimensional electrophoresis. The strip gel is separated from the slab gel by a fluid-impermeable and electrically insulative barrier. The barrier is preferably a wall which surrounds and forms a reservoir for containing the strip gel. A first dimension separation is carried out by pouring a liquid sample and a buffer solution into the reservoir to expand the gel, loading the sample into it, and feeding a current to the reservoir. The barrier which is easily breakably connected with the support is detached, so that the strip gel comes into contact with the slab gel to carry out a second dimension separation.

    Abstract: A strip gel and a slab gel are combined in a single gel enclosure or on a single gel backing for two-dimensional electrophoresis. The enclosure is comprised of a single pair of support plates, and a removable electrically insulating region is included to separate the gels. The region is occupied either by air or an electrically nonconductive material which can be removed without disturbing either gel. In use, a sample is loaded onto one end of the strip gel and an electric potential is imposed across the strip gel for the first dimension separation while the strip gel is electrically insulated from the slab gel. The insulating region is then removed and the two gels placed in electrical contact for the second dimension separation.

    Abstract: Polyacrylamide gels containing chaotropic agents such as urea as solubilizing agents for protein or nucleic acid separations are rendered stable for prolonged periods of time by the use of certain diacrylyl compounds with tertiary amide groups as cross linking agents. These compounds have the general structure where the R groups are generally saturated hydrocarbon groups and R through R may also be hydrogen. When used as cross linkers, these compounds have the further advantage of preventing urea crystallization at lowered temperatures.
