Abstract: PURPOSE: To obtain the provision of an enzyme immunoassay having a focus at the assay of vitamin B12 . CONSTITUTION: The denaturant used to pretreat a test sample for immunoassay is neutralized and inactivated by adding treating agent of the combination of acid buffer and reagent for converting the sulfhydryl group of the denaturant into inactivation type such as, for example, disulfide or alkylthio group. The treating agent is operated without denatured activity to disturb the sequential steps of this assay step. Further, the method for the assay of vitamin B12 using the pretreating agent is disclosed.
