The first chamber and the second chamber are separated by a membrane. The tube assembly further includes a spring positioned at the second end of the tube assembly. The tube assembly further includes a spring retainer configured to prevent the spring from decompressing when in a locked position and permit the spring to decompress when in an unlocked position.

In one aspect, described herein are methods of detecting the target nucleic acid.
