    Abstract: (A) Chemiluminescent 9-carboxy-acridinium derivs. of formula (I) are new, where R1 = H, 1-10C alkyl, alkenyl or alkynyl, benzyl or aryl; R2, R3 = H, 1-4C alkyl, opt. substd. amino, COOH, alkoxy, CN, NO2 or halogen; R4 = residue in which a sulphonamide group is linked via the N-atom with the carbonyl group, or a thio residue of the formula -S-X-R5; X = opt. branched aliphatic or azomatic group, opt. contg. a hetero atom; R5 = reactive group capable of reacting under mild conditions selectively with amino, carboxy, thiol or other functional groups in substances of biological interest; A = anion not adversely affecting the chemiluminescence. (B) Luminescent cpds. in which a cpd. (I) is bound directly or via a bridging molecule to a protein, polypeptide or other biologically interesting substance to form a stable, immunologically active complex are new and claimed. (C) Luminescence immunoassays for the determination of an antigenic substance in a liquid specimen in which at least one immunologically active component is immoblised on a solid phase in a competitive or sandwich process and in which the tracer is a luminescent cpd. as defined under (B) are new.

    Abstract: Determn. of the concn. of the free portion of an active ingredient (I) in a biological fluid in presence of its natural binder (II), the bound and free fractions of (I) being in equilibrium, comprises (1) treating the test sample with unlabelled antibody (Ab); (2) sepg. sample and Ab; (3) incubating Ab with a labelled, cross-reactive tracer (T), and (4) measuring amt. of T which is bound or unbound, and from this calculating the concn. of free (I). The novelty is that the amt. of Ab and/or its affinity to (I) is such that it does not significantly alter the equilibrium between free and bound forms, while T has a significantly higher or lower affinity for Ab than (I) itself. Also new is a test kit for the process. Specifically, Ab can be monoclonal or polyclonal, and T is labelled with a radioactive atom; fluoroescent or chemiluminescent gp.; enzyme or chromophore.
