Un proceso para producir un compuesto de la siguiente **fórmula** en la cual R1 representa hidrógeno, un grupo alquilo de 1 a 12 átomos de carbono, un grupo arilo de 6 a 12 átomos de carbono o un grupo aralquilo de 7 a 12 átomos de carbono, R4 y R5, independientemente, representan cada uno hidrógeno, un grupo alquilo de 1 a 12 átomos de carbono, un grupo arilo de 6 a 12 átomos de carbono o un grupo aralquilo de 7 a 12 átomos de carbono, y R4 y R5 pueden unirse entre sí para formar un anillo, que comprende: (1a) hacer reaccionar un enolato preparado permitiéndose que una base que contiene magnesio actúe sobre un derivado de éster acético de la siguiente fórmula X2CH2CO2R1 en la cual R1 representa hidrógeno, un grupo alquilo de 1 a 12 átomos de carbono, un grupo arilo de 6 a 12 átomos de carbono o un grupo aralquilo de 7 a 12 átomos de carbono, y X2 representa hidrógeno o un átomo de halógeno.

A process for the preparation of optically active 2-[6-(hydroxymethyl)-1,3-dioxan-4-yl]acetic acid derivatives, which comprises subjecting an enolate prepared by reacting an acetate ester derivative with either a base or a zero-valent metal to reaction with a hydroxybutyric acid derivative at -30 °C or above to thereby obtain a hydroxyoxohexanoic acid derivative, reducing this hydroxyoxohexanoic acid derivative with a microorganism into a dihydroxyhexanoic acid derivative, treating this dihydroxyhexanoic acid derivative with an acetal-forming reactant in the presence of an acid to thereby obtain a halomethyldioxanylacetic acid derivative, acyloxylating this halomethyldioxanylacetic acid derivative with an acyloxylating agent into an acyloxymethyldioxanylacetic acid derivative, and subjecting this acyloxymethyldioxanylacetic acid derivative to solvolysis in the presence of a base.

A process for efficiently producing optically active amino compounds, mainly (R)-amino compounds, at a low cost by means of a microbial enzyme; a polypeptide suitable for use in the production process and having a stereoselective transaminase activity; and a DNA coding for the polypeptide. The process is characterized by comprising causing a transaminase to act on a ketone compound as an amino acceptor in the presence of a primary amine as an amino donor to stereoselectively transfer the amino group to thus obtain an optically active amino compound. The DNA contains a base sequence coding for a polypeptide having a stereoselective transaminase activity. The polypeptide can be obtained by incubating a microorganism belonging to the genus Arthrobacter.

This invention provides a process for producing optically active 2-[6-(hydroxymethyl)-1,3-dioxan-4-yl]acetic acid derivatives, which are of value as intermediates of drugs, from inexpensive starting materials without using any special equipment such as that required for super-low temperature reactions. A process for producing optically active 2-[6-(hydroxymethyl)-1,3-dioxan-4-yl]acetic acid derivatives which comprises reacting a halomethyldioxanylacetic acid derivative with an acyloxylating agent to provide a acyloxymethyldioxanylacetic acid derivative, and subjecting this compound to solvolysis in the presence of a base.

A novel polypeptide for producing tert-butyl (3R,5S)-6-chloro-3,5-dihydroxyhexanoate, a gene encoding the polypeptide, and a method of using the polypeptide, are provided. The polypeptide has an enzyme activity to asymmetrically reduce tert-butyl (S)-6-chloro-5-hydroxy-3-oxohexanoate to tert-butyl (3R,5S)-6-chloro-3,5-dihydroxyhexanoate. The polypeptide comprises the amino acid sequence represented by SEQ ID NO. 1 in the Sequence Listing, or the amino acid sequence having at least one amino acid substitution, insertion, deletion, or addition thereof.
