Abstract: A dye composition for keratinous fibers which contains a coupler and a developer are disclosed. The present coupler component is a novel 2-alkoxy-3,5-diaminopyridine derivative of the formula (1): wherein R1 and R2 are the same or different from each other and independently represent a hydrogen atom, a lower alkanoyl group or a lower alkyl group which may be substituted by at least one hydroxyl group, and R3 is a lower alkyl group which may be substituted by at least one hydroxyl group, or a salt thereof; provided that R1 and R2 are not both a hydrogen atom or an acetyl group at the same time. The present 2-alkoxy-3,5-diaminopyridine derivatives are useful as couplers in dye compositions, and are capable of dyeing keratinous fibers such as hair in a bright blue color with excellent dyeability. Dye compositions containing the present compounds as a coupler show excellent resistance to discoloration, excellent resistance to shampooing and excellent resistance to friction.

Abstract: PURPOSE:To provide the subject compound useful as a coupling substance to be added to corneal fiber-dyeing compositions, and giving the composition capable of dyeing the corneal fibers such as hair into a blue color tone in a high chroma and excellent in the dyeability, discoloration resistance and washing resistance. CONSTITUTION:A compound of formula I (R , R are H, lower alkyl capable of being substituted with an OH group; Ar is substitutable aryl; m is 0-4; n is 1-4; l is 0, 1) or its salt, for example, 1,2-bis-(3,5-diamino-2-pyridyloxy)ethane.4 hydrochloride. The compound is obtained e.g. by reacting a 2-halogeno-3,5- dinitropyridine of formula II (X is halogen) with an aryl-substituted metal alkoxide of formula III (M is alkali metal; Ar' is Ar) in a solvent such as THF at a temperature of -5 deg.C to the boiling point of the solvent and subsequently reducing the product in the presence of a catalyst such as palladium carbon catalyst.

Abstract: PROBLEM TO BE SOLVED: To provide a method for testing the mutagenicity of a substance to be tested, capable of easily detecting the damage of DNA in high sensitivity by culturing an animal liver cell to which the substance to be tested has been dosed in the presence of a DNA-repairing agent, treating the nucleus fraction of the culture product with an alkali, and measuring the mol.wt. distribution of the DNA. SOLUTION: This method for testing the mutagenicity of a substance to be tested comprises extracting a liver from an animal such as a rat, transferring the liver into a buffer solution containing collagenase, finely cutting the liver with a knife, dispersing the obtained liver cells in a Hank's liquid cooled with ice, filtering the cells with a cell filter comprising folded gauze, washing the collected liver cells with Hank's liquid to obtain the lever cell. Then, the animal liver cells are dispersed in a culture medium, the substance to be tested is dosed to the culture medium, the liver cells are cultured in the presence of a DNA repair-inhibiting agent (e.g. aphidicolin) at 37 deg.C under the condition of 5% CO2 , a nucleus fraction from the obtained culture liver cells is collected, the collected nucleus fraction is treated with an alkali, and the mol.wt. distribution of the obtained single strand DNA is measured with the elusion speed from a filter. Thus, the DNA-damaging property of a carcinogenic substance can easily and highly sensitively be detected to test the mutagenicity of the substance to be tested.

Abstract: PURPOSE:To provide the subject derivative comprising a 2-alkoxy-3,5- diaminopyridine derivative, and useful as a coupling agent for corneal fiber- dyeing agents which can dye hair, etc., into a blue color tone having a high chroma and which have good dyeability, discoloration resistance and water resistance. CONSTITUTION:A 2-halogeno-3,5-dinitropyridine of formula I (X is halogen) (e.g. 2-chloro-3,5-dinitropyridine) is reacted with a metal alkoxide of formula MOR (R is lower alkyl which may have a hydroxyl group; M is alkali metal) (e.g. sodium methoxide) in methanol at room temperature, and the produced 2-alkoxy-3,5-dinitropyridine is reduced into a diamine derivative of formula II. The diamine derivative of formula II is mixed with acetic anhydride, etc., and a NaOH aqueous solution to give a pH of 6-7, and reacted with each other. The reaction product is diluten with water, extracted with a solvent and subsequently purified with a silica gel chromatography to obtain the 2-alkoxy-3,5- diaminopyridine derivative of formula III (R , R are H, lower alkanoyl, etc.).
