Abstract:
An object of the present invention is to provide: a novel method of analyzing diagnostic information, by which prostate cancer and a benign prostate disease can be distinguished from one another with high accuracy; and a diagnostic kit for obtaining the diagnostic information. One embodiment of the method of analyzing diagnostic information according to the present invention is a method of analyzing diagnostic information, which includes: measuring the concentration of a mucin-1 (Tn-MUC1), which is reactive with a lectin that recognizes and binds to an N -acetyl-D-galactosamine †’ serine (threonine) residue, in a blood sample originated from a subject to be diagnosed; comparing the thus measured value with a threshold value; and estimating that a disease affecting the subject to be diagnosed is not prostate cancer but a benign prostate disease when the measured value of the concentration of the mucin-1 is greater than the threshold value, and one embodiment of the diagnostic kit according to the present invention is a diagnostic kit including: an anti-mucin-1 antibody for quantifying Tn-MUC1; and a lectin for detection, which recognizes and binds to an N -acetyl-D-galactosamine †’ serine (threonine) residue.

[Problem] An object of the present invention is to provide a method by which the amount of a very small amount of analyte (in particular, one having a sugar chain) can be measured with a high accuracy and a device which is used for said measurement method. [Means for Solution] The method for measuring the amount of an analyte according to the present invention is characterized in that a lectin labeled with a fluorescent dye, preferably said lectin whose dissociation rate constant [k d ] is from 1.0 x 10 -6 to 1.0 x 10 -3 (S -1 ), is used as a secondary antibody in a sandwich assay using a surface plasmon excitation enhanced fluorescence spectroscopy [SPFS; Surface Plasmon-field enhanced Fluorescence Spectroscopy].

Provided are: an antigen detection method which can improve the detection sensitivity and quantitative performance for a sugar chain on an antigen to be detected with a lectin by a simple technique (simple constitution) in a sugar chain detection system using a lectin; and a detection kit. The antigen detection method of the present invention is a method of detecting an antigen having a specific sugar chain in a sample with a lectin that binds to plural kinds of sugar chains including the specific sugar chain, and the detection method comprises: a first step of bringing the lectin into contact with the sample; a second step of bringing a glycohydrolase capable of cleaving at least one kind of sugar chain to which the lectin can bind into contact with the sample, the at least one kind of sugar chain excluding the specific sugar chain among the plural kinds of sugar chains; and a fourth step of detecting the antigen bound with the lectin after the first and second steps.
