A monoclonal antibody specifically reactive to a human growth hormone with a molecular weight of about 20,000, a cell line capable of producing the monoclonal antibody and an immunoassay of a human growth hormone having a molecular weight of about 20,000 using the monoclonal antibody. An animal cell capable of producing an antibody specific to a human growth hormone having a molecular weight of 20,000 (20k hGH) was prepared from an animal immunized with 20k hGH. The animal cell was fused with a myeloma cell to produce a monoclonal antibody which specifically reacted to 20k hGH, but substantially not to 22k hGH. The monoclonal antibody was useful for immunoassay of 20k hGH.

Abstract:
PURPOSE:To provide a cell separator enabling stable high-density culture for a long period of time without filter cloggings. CONSTITUTION:The objective cell separator, which is installed out of a culture tank and independently thereof, is equipped with (A) a tank to hold culture fluid and cells and (B) a rotatable filter with its cross-section of closed design, having meshes 1 to 5 times the size of a carrier in the case cells to be cultured or such carrier is used. As the meshes are larger than the size of cells, cloggings do not occur. Rotation of the filter makes apparent meshes smaller, resulting in adequate separatory efficiency for cells. With this separator, stable cell separation can be continued for one month or longer.
Abstract:
PURPOSE:To recover a bioactive protein in a stable state, in production of the bioactive protein by perfusion culture of an animal cell. CONSTITUTION:pH of the recovered solution continuously obtained in perfusion culture of an animal cell is adjusted within a range of 6.0 to 7.0. Thereby, the bioactive protein can be prevented from being decomposed by a protease and the protein is stabilized.
Abstract:
PURPOSE:To obtain a synthetic medium enabling the production of valuable substances by animal cells without using protein components such as transferrin, insulin and albumin by using glycylglycine as an essential component. CONSTITUTION:This synthetic medium for the high-density culture of animal cells to produce valuable substances contains glycylglycine and is absolutely free from proteins acting as proliferation factor (e.g. transferrin, insulin and albumin). The amount of glycylglycine added to the medium is about 1-100mM, preferably about 5-15mM.
