Abstract: Apparatus for detecting light emitted by assay samples is provided, in which light emitted by the sample is collected for transmission to a charge coupled device camera (74) by an optical fibre bundle. The cross-sectional area of the optical fibre bundle corresponds to the area of the sample, the end of which is located close to the sample for detecting any light emitted therefrom, and selected fibres (30) of those making up the bundle are separated from the remainder and extend to a source of excitation radiation (76) and serve to convey excitation radiation (if required) directly to a corresponding plurality of points distributed over the area of the end face of the bundle and therefore over the area of the sample. The remaining fibres (32, 38) of the bundle serve to collect emitted light (whether generated by fluorescence caused by excitation or otherwise) and provide a light path to the charge coupled device camera, wherein the ends of the excitation fibres and the ends of the emitted light collecting fibres are distributed uniformly over the area of the fibre bundle presented to the reaction site.

Abstract: A method of detecting the presence and position of labelled material in a sample in which the labelled material either gives of light or can be stimulated to do so. The sample is imaged onto an image intensified CCD camera which is scanned following each exposure. Measurements are performed on the data signals so obtained, to identify clusters of data values from adjacent regions of the CCD array caused by light emitted by the image intensifier incident on those regions. The measured signal values are compared with at least one threshold so as to distinguish clusters resulting from light emitting regions of labelled material from the remainder of the sample and the centroid of each light produced cluster of data values is computed with reference to the camera array, and a signal value corresponding to the centroid coordinates is stored in a memory together with the centroid coordinates of any other light produced clusters identified during the same interrogation. The coordinates from each of a succession of interrogations of the same sample may be stored in an accumulation store to enable a list of the recorded light emissions and/or display of the events, to be produced by reading out the store. Apparatus for performing this method is also described.

Abstract: A method of detecting the presence and position of radio isotope material in a sample is described comprising the steps of: exposing the sample to a phosphor, repetitively imaging the phosphor onto an image intensified CCD camera, scanning the camera CCD array and interrogating the charge pattern thereon following each exposure, performing measurements on the data signals obtained thereby to identify clusters of data values from adjacent regions of the array caused by light emitted by the image intensifier onto those regions and comparing the measurements with threshold values and parameter values so as to distinguish clusters resulting from light emitted as a result of radioactive decay events energising the phosphor, from other light producing events, computing the centroid of each cluster of data values identified as a radioactive decay event with reference to the camera array and storing the centroid coordinates in a memory together with the centroid coordinates of any other radioactive decay produced clusters identified during the same interrogation. Coordinates from each of a succession of interrogations of the same sample are stored in an accumulation store, which can be read out to give a list of recorded events and/or used to produce a display of the events superimposed on an outline of the sample. The samples are carried on fibre optic plates in conctact with the photocathode and event coordinates are stored in memory as they are detected, so that the dispaly is updated as the events occur. Low noise bi-alkali material is used for the photocathode and the CCD camera is operated in inverted mode to reduce noise.
