Abstract: Procedimiento para producir una β-glucosidasa mutante derivada de un termófilo que tiene fijada selectivamente a ella una cadena de azúcares y también tiene una actividad glucosidasa, que comprende: (i) preparar ADN que codifica una β-glucosidasa mutante derivada de un termófilo introduciendo una secuencia de ADN que codifica Asn-X-Ser o Asn-X-Thr (en las que, X es cualquier aminoácido excepto prolina) en ADN que codifica una β-glucosidasa derivada de un termófilo que está originalmente desprovista de una secuencia de glucosilación y, además, añadiendo una secuencia de ADN que codifica una secuencia señal de secreción al ADN que codifica una β-glucosidasa mutante, (ii) introducir el ADN que codifica una β-glucosidasa mutante al que se le ha añadido la secuencia de ADN que codifica la secuencia señal de secreción en un microorganismo eucariota, de modo que una β-glucosidasa mutante codificada por el ADN de la β-glucosidasa mutante se exprese como una proteína secretora, y (iii) aislar y purificar la β-glucosidasa mutante expresada de este modo como una proteína secretora.

Abstract: Provided is an efficient method for decomposing biomass, which involves producing glucosidase in which the cellulose degrading efficiency is improved, and using said glucosidase. Specifically, provided is a method for producing a mutant glucosidase wherein a DNA encoding a secretion signal sequence and a DNA encoding Asn-X-Ser or Asn-X-Thr are introduced into a DNA encoding a glucosidase from a thermophile, and the resulting product is introduced into a eukaryotic microorganism and expressed as a secretory protein. Also provided are an enzyme composition containing said mutant glucosidase, and a biomass hydrolysis method using said enzyme composition.
