Abstract: In a fluorescence correlation spectroscopy process light (7) is focused on a measuring volume (16) into which a probe (12) bearing sample molecules (4) has been introduced. Light emitted by the sample molecules is measured by a detector (15). The novelty is that the molecules are distributed throughout the measuring volume in a suspension and that the molecules interact with metal particles. The metal particles are introduced into the measuring volume by an optical substrate (2). The incident stimulant light produces an intense lighted area in the immediate vicinity of the metal surface. This intensity amplifies the molecular fluorescent stimulation, greatly increasing the sensitivity of the measurement. Also claimed is a suitable fluorescence correlation spectroscopy microscope.
