    Abstract: A method of fraction collection suitable for a liquid chromatography system, comprising diverting a chromatography flow from a waste channel to a collection tube 38 at the start of a fraction collection window; collecting the chromatography flow dispensed from the collection tube during the fraction collection window in a collection vessel 28; diverting the chromatography flow to the waste channel at the end of the fraction collection window; and providing a flow of a wash solvent to the collection tube to dispense at least a portion of a liquid remaining in the collection tube at the end of the fraction collection window from the collection tube. Also disclosed is a fraction collector for a liquid chromatography system, comprising a valve 60 and four ports in fluidic communication with the chromatography flow, a waste channel, a wash solvent source 62 and a collection tube; the valve having a first state wherein the collection tube receives the chromatography flow and the wash solvent source is in fluidic communication with the waste channel and a second state wherein the collection tube receives a flow of wash solvent and the chromatography flow is in fluidic communication with the waste channel.

    Abstract: A dispensing needle 50 for a fraction collector comprising a conduit 52 defining a fluid channel 56 for conducting chromatography system flow, the fluid channel having a cross-sectional area; and a needle tip 54 having a fluid channel 58 and an endface 60, the fluid channel having a cross-sectional area that is less than that of the fluid channel of the conduit, the needle tip disposed at and in fluidic communication with the dispensing end of the conduit, thereby enabling a liquid chromatography system flow at a first velocity in the conduit to be dispensed at the endface of the needle tip at a second velocity that is greater than the first velocity. Preferably the conduit is flexible. The conduit and needle tip may be formed as a single integral body. Preferably the needle tip has a tapered outer surface, more preferably a truncated cone shape. The needle tip and conduit may be formed of metal, ceramic or glass. Preferably, at least one of the needle tip and the conduit is formed of titanium, titanium alloy or fused silica.

    Abstract: A dispensing needle 50 for a fraction collector, comprising a conduit having a fluid channel 58 to conduct a chromatographic flow, an interior wall defining the fluid channel, an exterior surface 64 and an endface 60 through which the chromatographic flow is dispensed; and a coating 62 of a hydrocarbon material covalently bonded to the endface, to at least a portion of the exterior surface and to at least a portion of the interior wall, the portions of the exterior surface and the interior wall being adjacent to the endface, wherein the coating of a hydrocarbon material operates to reduce a droplet volume of a liquid dispensed from the endface. The hydrocarbon material is preferably a self assembled monolayer of a hydrocarbon material and preferably bonded by a monopodal attachment. The hydrocarbon material may have a carbon chain length of at least six and is preferably less than the carbon chain length of a stationary phase in the chromatographic flow. Also disclosed is a dispensing needle comprising a coating of fluorocarbon material covalently bonded to the endface, to at least a portion of the exterior surface and to at least a portion of the interior wall.
