Abstract: A method comprising obtaining a porous medium, frit or plug 312a’ comprising a porous material having a first shape and an initial porosity profile, and engaging the porous medium with a cavity 310’ in a microfluidic device wherein the cavity is in fluid communication with a channel 306’ of the microfluidic device and engaging the porous medium with the cavity causes the first shape of the porous material to be adjusted to a second shape according to a shape of the cavity which results in the initial porosity profile being adjusted to a target porosity profile according to the shape of the cavity, the target porosity profile being uniform. Ideally, the porous material is compressed and deforms to the shape of the cavity to achieve the target porosity profile. Preferably, the cavity is in fluid communication with the channel via an opening of the channel which is an inlet of the channel. The porous material may comprise sintered particles.

Abstract: An apparatus comprising a fluidic device 300 having a fluidic channel 308 and a cavity 310, the fluidic channel having a channel diameter to accommodate a fluid flow and the cavity having an open end in proximity to a surface of the fluidic device, wherein the cavity has a dimension that is greater than the channel diameter and is in fluid communication with the fluidic channel via a channel opening 306. A preformed porous medium 312a comprising a porous material is disposed within the cavity, the porous medium having a porosity profile and being in fluid communication with the fluidic channel via the opening, wherein fluid flow is directed through at least a portion of the porous medium. The porosity profile comprises an inner region having a first porosity and an outer region having a second porosity that differs from the first porosity, wherein the inner region and the outer region are aligned along a common axial reference location. Ideally, the fluidic device is a microfluidic device. Preferably, the first porosity is greater than the second porosity and extends through the porous medium such that fluid flow is directed through the inner region of the porous medium.

Abstract: A method comprising obtaining a porous medium 412 comprising a porous material having a first shape and an initial porosity, engaging the porous medium with a cavity 418 in a fluidic device, the cavity being in fluid communication with a channel 416 of the device, wherein the engagement of the porous medium with the cavity causes the first shape of the porous medium to be adjusted to a second shape resulting in the initial porosity being adjusted to a second porosity. Ideally, the porous material is a frit or retaining plug in a separation column such as a planar microfluidic device. The frit maybe compressed and ideally the porosity of the outer portion 424 of the frit is reduced whilst the inner porosity 426 remains unchanged. Peak broadening is reduced. A device comprising the porous medium is also disclosed.
