Abstract: A receptacle cap includes a probe recess configured to receive a probe inserted therein to thereby frictionally secure the receptacle cap to the probe, and a closed lower end of the receptacle is insertable into an open end of a reaction receptacle to close the reaction receptacle. The receptacle cap and the reaction receptacle include interlocking features for securing the receptacle cap to the reaction receptacle.

Abstract: A lyophilization nest and method of using the same is described herein. In various embodiments, the lyophilization nest includes a base and first and second covers and is configured to support one or more receptacles each supporting one or more substances within interior spaces of the lyophilization nest. The interior spaces may be in fluid communication with the exterior of the lyophilization nest through one or more vent holes extending through the first and second covers. Each of the one or more vent holes have a corresponding sealing element configured to selectively form an air-tight seal within the vent holes, such that a controlled environment may be maintained within the interior spaces when the ambient conditions surrounding the lyophilization nest are not lyophilization conditions. The one or more sealing elements may be operable while the lyophilization nest is positioned within a sealed lyophilizer by depressing the sealing elements into corresponding vent holes to form the air-tight seal.

Abstract: A method for performing a nucleic acid amplification assay employs a thermally-conductive receptacle holder with one or more receptacle wells, each conforming to an outer surface of a lower portion of a vial. A through-hole extends from an inner surface of each well to an outer surface of the holder. A thermal element is positioned proximal to the holder for altering a temperature of the holder. A signal detection module is configured to generate an excitation signal directed through the through-hole of the well and detect an optical emission from a fluid contained in the vial supported by the well. At least one well supports a capped vial containing a reagent for a nucleic acid amplification assay and including an opaque cap sealing an open end of the vial. The lower portion of the vial is contained within a well, and the cap is situated above a top surface of the holder.

Abstract: An apparatus for separating an interlocked cap and receptacle comprises a top wall and a cap removal station. The cap removal station comprises an opening, a raised collar for engaging and releasing interlocking elements of the cap and receptacle, and tabs on a side of the top wall opposite the raised collar for retaining the receptacle within the opening when the cap is separated from the receptacle. A method for separating an interlocked cap and receptacle comprises moving the cap and receptacle in a first direction into a cap removal station, contacting interlocking elements of the cap and receptacle with a raised collar to thereby release the interlocking elements, retaining the receptacle within the cap removal station; and with the interlocking elements released and the receptacle retained within the cap removal station, moving the cap in a second direction opposite the first direction to separate the cap from the receptacle.

Abstract: An apparatus configured for mixing the contents of one or more fluid containers includes a fluid container support platform configured to hold one or more fluid containers. The fluid container support platform is configured to index the container to one or more specified locations and to be moved in an orbital path about an orbital center independently of the rotation about the central axis of rotation. The apparatus further includes an indexing drive system configured to effect indexing movement of the container support platform and a vortex drive system configured to effect powered orbital movement of the container support platform about the orbital center. An evaporation limiting insert placed within containers reduces exposure of the fluid contents of the container to atmospheric air, thereby reducing susceptibility of the fluid contents to evaporation.

Abstract: A method for reconstituting a lyophilized reagent contained within a reagent well comprises the steps of drawing a diluent into a pipette tip attached to an automated pipettor and dispensing the diluent into the reagent well containing the lyophilized reagent. The reagent well has an internal side wall, a bottom wall, and an open upper end and includes one or more retention features disposed about the periphery of the internal side wall and defining a central opening into the well that permits passage of the pipette tip into the reagent well. The one or more retention features are integrally formed with the internal side wall, and each of the one or more retention features extends over a portion of the lyophilized reagent, thereby retaining the lyophilized reagent within the reagent well.

Abstract: A cap that is securable to a vial includes a plug configured for insertion into an open end of the vial. An upper portion of the cap defines a probe recess with an open top end and configured to receive a distal end of a probe of a pipettor inserted into the open top end, to frictionally secure the cap to an end of the probe. The cap includes a radially-oriented annular surface with one or more locking members depending from a periphery of the annular surface. The locking members are spaced from the plug and each locking member includes a radial locking groove and a radial locking ridge beneath the radial locking groove. A lip of the vial snaps into the radial locking groove above the radial locking ridge to secure the cap to the vial when the plug is inserted into the open end of the vial.

Abstract: Receptacles containing a reaction mixture are transported to receptacle wells of a thermally conductive receptacle holder, and the temperature of the holder is cycled with a thermoelectric device situated between a support and a first side of the holder while a first force is applied onto a second side of the holder. Optical communication between each well and an excitation signal source and an emission signal detector is established to determine whether an emission signal is emitted from any of the receptacles during temperature cycling as an indication of the presence of a target nucleic acid. The thermoelectric device may be situated between an upright portion of the support and the first side of the holder. A second force may be applied onto a top end of each receptacle in the holder by a cover moveable with respect to the receptacles between an open position and a closed position.
