A system and method for automated processing of nucleic acids and other samples includes a disposable container comprising a tray and a flexible barrier. The barrier is configured to seal with a top edge of the tray, providing a closed, aseptic work area within the sealed tray. A pipette head and/or other sample manipulation device can be attached to the inside of the barrier, and the barrier can include an interface for a robotic arm or other device. When the barrier is sealed over the tray, the barrier separates the contents of the tray from the robot or other manipulation device. The barrier can be flexible, and allow the robotic arm to move the pipette head throughout the work area of the tray. All samples, reagents, pipette tips and other tools or devices for processing nucleic acid samples may remain within the closed compartment provided by the container during processing.

An isolator includes a work chamber, a sterilizing substance supply unit, a gas flow channel pressure adjustment unit, a work chamber barometer, and a controller. The controller is configured to control execution of a gas flow channel leak test for checking a gas leak in a gas flow channel based on a detection result by the work chamber barometer after making the gas flow channel pressure adjustment unit increase or decrease the pressure in the gas flow channel, and is configured to control supply of the sterilizing substance by the sterilizing substance supply unit. The controller performs heating of a heater, which accompanies the supply of the sterilizing substance, in parallel with the gas flow channel leak test.

An apparatus and a process for forming a protective barrier seal along a “ring of concern” of a transfer container used with double door systems is provided. A protective substrate is supplied between a “ring of concern” and a safety cover in which an adhesive layer of the substrate engages the “ring of concern”. A compressive foam strip along an opposite side of the substrate engages a safety cover such that a compressive force is maintained between the “ring of concern” and the adhesive layer of the substrate.

The present invention provides a device for collection of liquid samples under atmospherically-isolated conditions, the device including a hermetically-sealable chamber having one or more sample manipulating members or similar equipment within the chamber, and a pump system for drawing liquid through a sampling tube into the chamber directly or into a container. The container includes an outlet within the chamber for dispensing the collected sample under atmospherically-isolated conditions. Also provided is a device wherein the pump system that includes an outlet within the chamber for dispensing collected liquid sample.
