Disclosed herein is a method and apparatus for gel electrophoresis which employs periodic inversion of the electric field essentially in one dimension. According to this system, which is defined herein as field-inversion gel electrophoresis, net migration is achieved by using a longer time or higher voltage in one direction than in the other direction.

A method and apparatus for gel electrophoresis which employs periodic inversion of the electric field essentially in one dimension, denoted as field-inversion gel electrophoresis (FIGE), results in net migration by using a longer time or higher voltage in one direction than in the opposite direction. FIGE permits separation of DNA or protein mixtures in size ranges not accessible to ordinary electrophoresis.

A high precision, small volume fluid processing system employs open ended capillary tubes to meter, aliquot and mix small volumes of sample fluid and reagents. The system has an automatic mechanism for moving the capillary tubes as well as automated sub-systems for incubating and mixing fluids within the capillary tubes.
Abstract:
A large DNA cloning system is disclosed which is based on yeast artificial chromosomes. Cloning vectors are disclosed which allow the cloning of large segments of greater than 50 kb of exogenous DNA. The cloning vector comprises DNA sequences of an autonomous replication sequence, a centromere, a selectable yeast marker, two sequences that seed telomere function in vivo, and a cloning site within an interruptible yeast gene for insertion of the exogenous DNA segments.
