Abstract: A method for identifying Korean native cattle is provided to distinguish the Korean native cattle from imported beef with excellent accuracy by using microsatellite loci, thereby improving income of a farmhouse raising the Korean native cattle and the reliability of consumers. The method comprises the steps of: (a) preparing a PCR reaction solution for amplifying alleles corresponding to microsatellite loci of a selected cow such as ILSTS005, ILSTS028, ILSTS103, ILSTS006, ILSTS008, ILSTS023, ILSTS050, TGLA122, TGLA126, MGTG4B, TGLA227, BM1824, BM2112, ETH10, ETH225, ETH3, and SPS115; (b) amplifying the alleles by performing the multiple PCR reaction thereon using a PCR reaction solution which is a combination of ETH225, ETH10, ETH3, BM2113, BM1824 and SPS115; (c) isolating each of the amplified alleles and then analyzing the size of the each of the alleles; (d) putting the size of the alleles by individuals and species in order and preparing the number and the frequency distribution of the alleles by the species based thereon; and (e) building an information bank of the alleles corresponding to the microsatellite loci to identify the species according to the statistical analysis.

Abstract: A DNA marker is provided to determine an individual genetic trait of Korean native cattle by using genotypes of three independent loci of a FABP4(adipocyte-fatty acid binding protein) gene which has the significantly effect for carcass weight and marbling of the Korean native cattle, thereby being utilized for an early selection of a fatting beef and an accurate estimate of breeding values. A method for selecting a DNA marker of determining genetic traits comprises the steps of: (a) identifying a genotype of a linkage group detected from a Korean native cattle a fatty acid binding protein(FABP4) gene of SEQ ID : NO. 7 using the decoding of 17 SNP loci depicted at a Fig. 1; (b) after pre-denaturing the Korean native cattle FABP4 gene of SEQ ID : NO. 7 as a template using a PCR primer of SEQ ID : NOs. 1 to 6 at a temperature of 94 deg.C for 5 minutes, performing 35 times of a cycle where one minute annealing at a temperature of 58 deg.C and two-minute expanding at a temperature of 72 deg.C are done and then finally reacting it for 10 minutes at a temperature of 72 deg.C for PCR amplification; (c) after cleaving an amplification product obtained from the step(b) with an restriction enzyme such as Hpy188I, RsaI and NlaIII, subjecting it to an electrophoresis; and (d) obtaining a picture and an image of the electrophoresis product to determine an individual genotype through a restriction fragment length polymorphism analysis, a heteroduplex analysis, a single strand conformational polymorphism(SSCP) and a Taqman probe method. A DNA marker is selected by the method and constructed to recognize three loci of Hpy188I(C->G), RsaI(A->T) and NlaIII(A->G) described in the Fig.1 among variable regions of a FAB4 gene expressed in adipocyte.

Abstract: A method for identifying Korean native cattle and imported beef is provided to accurately identify the Korean native cattle derived from Bos taurus and the imported beef derived from Bos indicus by using structural differences of an RFLP genotype of a sex-determining region Y(SRY) gene. The method comprises the steps of: (a) collecting DNA from Korean native cattle and imported beef respectively; (b) respectively combining a forward primer having a nucleotide sequence of 5'-TTAGAACGCTTACACCGCATATTACTTC-3' and a backward primer having a nucleotide sequence of 5'-TAGTAAAATTGAGATAAAGAGCGCCTTTGTTAG-3' of an SRY gene with each of the collected DNA; (c) amplifying the combined product through PCR in which a nucleotide sequence portion of 1003-1803 in a Bos taurus DNA(registration number : AB039748) and a nucleotide sequence portion of 273-1073 of a Bos indicus DNA(registration number : AY079145) are amplified; and (d) after cutting the amplified product with an MseI restriction enzyme, analyzing polymorphism thereof to divide genotypes of the Korean native cattle and the imported beef.
