Abstract: Human antibodies, preferably recombinant human antibodies, that specifically bind to human tumor necrosis factor α (hTNFα) are disclosed. These antibodies have high affinity for hTNFα (e.g., Kd=10−8 M or less), a slow off rate for hTNFα dissociation (e.g., Koff=10−3 sec−1 or less) and neutralize hTNFα activity in vitro and in vivo. An antibody of the invention can be a full-length antibody or an antigen-binding portion thereof. The antibodies, or antibody portions, of the invention are useful for detecting hTNFα and for inhibiting hTNFα activity, e.g., in a human subject suffering from a disorder in which hTNFα activity is detrimental. Nucleic acids, vectors and host cells for expressing the recombinant human antibodies of the invention, and methods of synthesizing the recombinant human antibodies, are also encompassed by the invention.

Abstract: Un método para obtener un anticuerpo de especificidad dual o parte de unión al antígeno del mismo, que se une específicamente a dos proteínas diferentes pero relacionadas estructuralmente, comprendiendo el método: proporcionar un antígeno diseñado sobre la base del corte y empalme de partes solapantes entre sí de las dos proteínas diferentes pero relacionadas estructuralmente para crear un péptido híbrido, donde dicho péptido híbrido tiene una estructura X-Y-Z, en la que Y representa una región de identidad o de fuerte similitud entre las dos proteínas relacionadas, X representa una región de una de las proteínas relacionadas y Z representa una región de la otra de las proteínas relacionadas; exponer un repertorio de anticuerpos al antígeno; y seleccionar del repertorio un anticuerpo que se una específicamente a las dos proteínas diferentes pero relacionadas estructuralmente para obtener de esta manera el anticuerpo de especificidad dual.

Abstract: Invenţia se referă la un anticorp recombinant uman, care se leagă la factorul de necroză umană alpha (hTNF alpha ) care are afinitate pentru hTNFalpha, Kd = 10-8M sau mai mică, şi o constantă a vitezei Koff de 1 x 10-3s-1 sau mai mică, şi neutralizează activitatea in vitro şi in vivo, precum şi la un acid nucleic care îl codifică, celula gazdă care cuprinde un vector de expresie recombinant, precum şi la o compoziţie farmaceutică ce cuprinde anticorpul, administrată în tratamentul bolilor în care activitatea TNFalfa este dăunătoare.
