Abstract:
PROBLEM TO BE SOLVED: To provide antibodies having dual specificity for two different but structurally related antigens.SOLUTION: The antibodies have dual specificity for IL-1α and IL-1β and neutralize IL-1α and IL-1β activity in vitro and in vivo. The antibody is a full-length antibody or an antigen-binding portion thereof. Methods of making and methods of using the antibodies are also provided.
Abstract:
Antibodies having dual specificity for two different but structurally related antigens are provided. The antibodies can be, for example, entirely human antibodies, recombinant antibodies, or monoclonal antibodies. Preferred antibodies have dual specificity for IL-1 alpha and IL-1 beta and neutralize IL-1 alpha and IL-1 beta activity in vitro and in vivo . An antibody of the inventi on can be a full-length antibody or an antigen-binding portion thereof. Methods of making and methods of using the antibodies of the invention are also provided. The antibodies, or antibody portions, of the invention are useful for detecting two different but structurally related antigens (e.g., IL-1 alpha and IL-1 beta ) and for inhibiting the activity of the antigens, (e.g., in a human subject suffering from a disorder in which IL-1 alpha and/or IL-1 beta activity is detrimental.

Abstract:
A dual-specificity antibody or antigen-binding portion thereof specifically binds interleukin-1alpha and interleukin-1beta wherein the antibody is not a fully mouse antibody. The antibody can be used in diagnostic methods for detecting IL-á or IL-â and in the manufacture of a medicament for treating interlauikin-1-related disorders such as inflammatory disorder, autoimmune disorder i.e. rheumatoid arthritis, Crohn's disease, multiple sclerosis, insulin dependant diabetes mellitus and psoriasis. Methods of obtaining dual-specificity antibodies and methods of making an antibody are also described.

Abstract:
Human antibodies, preferably recombinant human antibodies, that specifically bind to human tumor necrosis factor 'alpha' (hTNF'alpha') are disclosed. These antibodies have high affinity for hTNF'alpha'(e.g., Kd = 10exp(-8) M or less), a slow off rate for hTNF'alpha' dissociation (e.g., Koff = 10exp(-3) sec-1 or less) and neutralize hTNF'alpha' activity in vitro and in vivo. An antibody of the invention can be a full-length antibody or an antigen-binding portion thereof. The antibodies, or antibody portions, of the invention are useful for detecting hTNF'alpha' and for inhibiting hTNF'alpha' activity, e.g., in a human subject suffering from a disorder in which hTNF'alpha' activity is detrimental. Nucleic acids, vectors and host cells for expressing the recombinant human antibodies of the invention, and methods of synthesizing the recombinant human antibodies, are also encompassed by the invention.
