Abstract:
PURPOSE: A recombinant swine influenza A virus and a vaccine containing the same are provided to suppress proliferation of the virus and to prevent domestic pigs from the virus. CONSTITUTION: A recombinant swine influenza A virus contains gene of sequence numbers 1-6 among internal genes of A/swine/Korea/CAN01/04 (H1N1) of deposit number KCTC11165BP and virus surface protein gene of deposit number KCTC 11691BP. The virus surface protein gene contains HA of sequence number 7 and NA gene of sequence number 8. A vaccine for swine influenza virus contains the recombinant swine influenza A virus.
Abstract:
A mixed live vaccine of transmissible gastroenteritis virus, porcine epidemic diarrhea virus and porcine rotavirus, and a vaccination method by using the same live vaccine are provided to prevent diarrhea of pigs at one time, reduce vaccination time, and minimize titer loss of attenuated viruses by using trehalose. The mixed live vaccine is prepared by mixing a transmissible gastroenteritis virus(KCTC 10889BP), a porcine epidemic diarrhea virus(KCTC 10891BP) and a porcine rotavirus(KCTC 10890BP) in a weight ratio of 1:1:1, and adding a trehalose phosphate glutamate gelatin mixture. The mixed live vaccine is vaccinated by vaccinating a mother pig with the mixed live vaccine one time, and vaccinating the first vaccinated mother pig with the vaccine of transmissible gastroenteritis virus(KCTC 10889BP) and porcine epidemic diarrhea virus(KCTC 10891BP) two times.

본 발명은 불활화시킨 TGEV 및 PEDV를 항원으로 하는 혼합불활화 백신을 제공한다. 또한, 본 발명은 (1) TGEV를 ST세포에서 배양하고, PEDV를 Vero 세포에서 배양하는 단계; (2) 상기 단계 1의 배양액을 원심분리하여 고역가의 TGEV 및 PEDV (10 7.0 TCID 50 /ml 이상)를 함유한 상층액을 분리하는 단계; (3) 상기 단계 2에서 얻은 상층액에 바이러스 불활화제를 첨가하여 각 바이러스를 불활화시키는 단계; (4) 상기 불활화시킨 배양액을 ST 및 Vero 세포에 접종하여 세포변성유무를 확인하는 단계; 및 (5) 상기 불화화시킨 바이러스 배양액을 혼합하는 단계를 포함함을 특징으로 하는 혼합불활화 백신의 제조방법을 제공한다. 또한, 본 발명은 상기 본 발명에 따른 혼합불활화 백신을 돼지의 분만 5∼7주전 및 2∼3주전에 각각 2회 접종하여 모돈의 초유로부터 TGEV 및 PEDV 항체를 자돈에 전달하는 것을 특징으로 하는 돼지바이러스성 설사병의 예방방법을 제공한다. 상기 구성에 의하면, 주요 돼지 바이러스성 설사병의 원인체인 TGEV 및 PEDV를 충분히 예방하여 안전성이 우수하면서도 포유자돈에서의 설사병으로 인한 피해를 최소화하는 것이 가능해진다.

PURPOSE: A PAM-pCD163 cell line for isolating porcine reproductive and respiratory syndrome(PRRS) virus is provided to develop a vaccine and to prevent PRRS. CONSTITUTION: A PAM-pCD163 cell line is prepared by expressing pCD163 in PAM cells through a retrovirus prepared by transforming plasmid containing pCD163 genes into HEK293 cells. The plasmid is pFB-Zeo-pCD163. A method for preparing the cell line comprises: a step of performing transfection of VSV-G glycoprotein and MMLVgag/pol gene, and a plasmid containing pCD163 into HEK293 cells to prepare retrovirus; and a step of inoculating the retrovirus into the PAM cells.
Abstract:
PURPOSE: A scFv gene of monoclonal antibody 4G31 and a recombinant protein expressing the same are provided to neutralize RAV and to reduce and diagnose rabies virus. CONSTITUTION: A 4G31 scFv(single chain variable fragment antibody) gene which specifically neutralizes rabies virus has an amino acid sequence and a base of sequence number 16. The 4G31 scFv gene is a 4G31 monoclonal antibody containing a gene encoding a heavy chain variable region(VH) and a light chain variable region(VL). An expression vector expresses the 4G31 scFv gene. A recombinant protein is prepared by expressingthe 4G31 scFv gene in E.coli using the expression vector. A therapeutic agent for rabies virus contains the recombinant protein expressed from the 4G31 scFv gene.

A kit and a method for diagnosing a porcine epidemic diarrhea virus antigen are provided to diagnose the antigen conveniently and rapidly without a special testing equipment in a farm or a laboratory field. A kit for diagnosing a porcine epidemic diarrhea virus antigen is characterized in that a conjugate of a monoclonal antibody 6G15(deposition no. KCTC 10996BP) coupled to nucleocapside 58kDa of an epidemic diarrhea virus and a gold particle is attached to a reaction position of the porcine epidemic diarrhea virus antigen on a strip and a monoclonal antibody 9D11(deposition no. KCTC 10997BP) is attached to a position different from the coupling portion of the porcine epidemic diarrhea virus antigen and the monoclonal antibody 6G15. A method for diagnosing the porcine epidemic diarrhea virus antigen comprises the steps of: (a) loading a sample onto a detection pad on a strip; (b) reacting the conjugate of the monoclonal antibody 6G15 coupled to nucleocapside 58kDa of the epidemic diarrhea virus and the gold particle with the porcine epidemic diarrhea virus antigen in the sample; and (c) reacting the porcine epidemic diarrhea virus antigen, the monoclonal antibody 9D11 and the reaction product obtained from the step(b).
