Abstract: A blood separation system for separating mono nuclear cells from whole blood is described. The system comprises a chamber (12) that is rotatable about a rotational axis (28). The chamber comprises first and second spaced apart walls (24, 26) that form a separation zone having a high-G side and a low-G side which is located closer to the rotational axis than the high-G side. The chamber also includes an inlet region (74) where whole blood enters the separation zone for separation into red blood cells (96), a plasma constituent carrying platelets (98), and an interface (58) carrying mono nuclear cells between the red blood cells and the plasma constituent. A plasma constituent collection region (76) is also provided in the chamber where the plasma constituent is collected for removal from the chamber. An outlet path (46, 206) communicates with the plasma constituent collection region (76) and provides for removal of the interface (58) from the chamber. The system is operated under the control of a controller (134) that is operable in first and second modes. In the first mode, whole blood is conveyed into the inlet region while red blood cells and the plasma constituent are removed from the chamber and while the interface (58) is maintained at a set location within the chamber. In the second mode, the controller operates to move the interface from the set location for removal from the chamber via the outlet path. Finally, the system is equipped with a sensing element (226) for detecting mono nuclear cells in the interface in the outlet path. A method of separating mono nuclear cells from whole blood is also described.

Abstract: Systems and methods rotate a chamber (14), in which whole blood is centrifugally separated into an interface between the packed red blood cells and the plasma constituent. The interface carries platelets and mononuclear cells. The systems and methods include an interface control unit which is operative in a first state to enable removal of platelet-poor plasma (PPP) in a first container for use as a diluting liquid, and in a second state, to retain mononuclear cells (MNC) in the chamber (14), while removing plateletrich plasma (PRP) from the chamber (14), bypassing the platelet-poor collection container, thereby maintaining its platelet-poor character, and in a third state to enable the removal of mononuclear cells (MNC) from the chamber (14) in a path (T13) that leads to a second container, where the mononuclear cells are collected.

Abstract: A blood separation system operates in a first mode to convey whole blood into the inlet region (48) of a blood processing chamber (38) for centrifugal separation into packed red blood cells, a plasma constituent, and an interface, which carries mononuclear cells, between the packed red blood cells and the plasma constituent. The system removes packed red blood cells and the plasma constituent from the chamber (38), while maintaining the interface within the chamber (38). The system operates in a second mode to remove the interface from the chamber (38) by conveying packed red blood cells into the inlet region (48). An outlet path (46) conveys the removed interface from the chamber (38). The outlet path (46) includes a first sensing element (OS) to locate mononuclear cells in the removed interface and provide a sensed output upon locating mononuclear cells.

Abstract: Systems and methods for separating mono nuclear cells from whole blood rotate a separation compartment (38) about a rotational axis. Whole blood is introduced from an inlet region (74) into a first region (76) of the chamber during its rotation to separte the whole blood into a plasma constituent, red blood cells and interphase between the red blood cells and the plasma constituent. The interface contains mono nuclear cells. While conveying whole blood into the first region, the plasma constituent is collected in the first region, while collecting the red blood cells in a second region of the chamber spaced away from the inlet region. Red blood cells are than conveyed into the second region (78) in a back flow direction toward the inlet region. While conveying red blood cells into the second region, the mono nuclear cells are collected in the first region.
