    Abstract: Macromolecular species in a sample are chromatographically separated in a manner analogous to that which would occur in molecular sieve chromatography (gel filtration), by being passed through a separation medium comprised of a non-water-soluble fluid-permeable plug of cross-linked polacrylamide, where the degree of crosslinking is sufficient to cause discontinuities, gaps or channels in the polymer network large enough to permit water and the macromolecules in the sample to pass through, and yet interfering with the free flow of the macromolecules sufficiently to cause their bulk flow rate through the plug to vary with their molecular weights. By varying the nature and composition of the monomers, the plug can be designed alternatively for a wide variety of types of chromatography, including ion exchange, hydrophobic interaction, affinity, boronate and dye-ligand chromatography.

    Abstract: Multiple electropherograms or chromatograms representing successive stages of an electrophoretic or chromatographic separation in a capillary are obtained by training a recorder on a succession of detection windows in the capillary, spaced apart along the capillary length. A single immovable detector is used, and the capillary is either held such that each detection window is placed in alignment with the detector in succession, or is looped such that the detection windows all lie in the path of the detector at the same time.

    Abstract: ELECTROPHORETIC SIEVING IN GEL-FREE MEDIA WITH DISSOLVED POLYMERS Separation of sample ions on the basis of molecular size is achieved by electrophoresis in an aqueous solution containing a dissolved non-crosslinked polymer. The polymer has a molecular weight range which overlaps that of the sample ions being separated. Species which vary in molecular weight but not in charge/mass ratio are separated by this method, which is of particular interest in high performance electrophoresis in capillary columns where the use of gels would be awkward and inconvenient.

    Abstract: Preparation of a separation medium for continuous bed reversed-phase chromatography comprises: (a) forming a mixture composed of (i) a water-soluble monomer selected from vinyl, and (meth)acrylic monomers, (ii) a crosslinking agent, (iii) an alkylmethacrylate in which the alkyl group has 3-30C, and (iv) a detergent, in an aqueous medium; and (b) polymerising (i), (ii), and (iii) in a one-step reaction. Also claimed is a preparation in which the mixture is composed of (i), (ii), (iii) a water-soluble monomer having a terminal vinyl group and a charged group, (iv) an alkyl methacrylate as described, and (v) a detergent as described, with polymerisation of (i), (ii), (iii), and (iv) taking place in a one-step reaction.

    Abstract: Bulk fluid movement in a capillary electrophoresis system is achieved by imposing a temperature change on either a portion of the capillary or a bulb joined to the capillary at one end. The process may be used for sample loading, in which case a small volume introduction is made, or for flushing of the capillary with buffer solution, in which a larger volume introduction is made. The temperature change may be either an increase or a decrease, and the volume passed into or through the capillary is controlled to a high degree of precision by selection and accurate measurement of the volume of the bulb or capillary portion to which the temperature change is applied together with accurate control of the temperature change itself.
